Objective:To study the function of Sanjinpian(SJP) on chronic bacterial prostatitis in rats and to study the mechanism of SJP to provide pharmacological basis for more extensive application of SJP in clinic.Methods:70Wistar male rats of250±20g were randomly divided into six groups, including the control group with10rats, model control group, nofloxacin group, high, medium and low dose of SJP group with12rats respectively. Rats of all groups except the control group were modeled by injection of E. coli bacteria suspension and performing operation. The control group was taken as the negative control, while the nofloxacin group was taken as positive control. On the7th day after making the model, the rats were given drugs, according to their weight. The administration dosage of SJP of high, medium and low dose group were10.50g/kg,5.25g/kg,2.63g/kg respectively,(The dose was converted as20,10,5times of therapeutic dose in clinic, respectively). Nofloxacin dosage was0.14g/kg (10times of usual clinical dose). Control group and model control group were given the same amount of distilled water. The drugs or distilled water were given to the rats everyday, and the weights of the rats were recorded every7days. Samples were collected and the index was evaluated for all groups21days after the administration.(1) The rats of each group were anaesthetized with10%chloral hydrate. Then the prostatic massage liquid (EPS) was taken and placed in glass slides. EPS pH value was measured. The small bulk density of lecithin and the number of white blood cells (WBC) were observed under the microscope.(2) Each rat was sacrificed after weighting. The prostate tissue was exercised and weighted. Part of the prostate tissue was taken, fixed in10%formalin solution as histological specimens and observed under the microscope to see the pathological changes of the prostate tissue.(3) Each rat was sacrificed and the prostate tissue was exercised. Physiological saline was added to the tissue to make10%prostate tissue homogenate. After centrifugation, supernatant fluid was collected and the content of SIgA, IL-1β and TNF-α in the supernatant fluid were measured respectively by enzyme linked immune sorbert assay(ELISA) kit.(4) The rats were sacrificed and the prostate tissue was exercised to make10%prostate tissue homogenate. The content of NO, Zn2+, NOS were measured by colorimetry.(5) Inhibition experiment in vitro:Microdilution method and Paper disc agar diffusion method were used in order to evaluate the inhibition bacteria effect of SJP.Results:(1) After observation of prostate massage fluid message of smear under a microscope, after given drugs for21days, compared with model group, the Leukocyte number of rats in SJP of high, medium and low dose group significantly reduced, and the lecithin density and the pH value of prostatic massage liquid returned normal.(2) According to the results of the pathological observation, after given drugs for21days, rats in SJP of high, medium and low dose group had obvious reduced in epithelial cells degeneration necrosis degree, interstitial congestion edema degree, inflammatory cells infiltrating degree than that of the control group.(3) The result of prostate homogenate biochemical index determination showed that SJP could improve the content of SIgA, Zn2+of rats with chronic bacterial prostatitis, and the NO, IL-1β, TNF-acontent in prostate tissue.(4) Inhibition experiment in vitro showed that the drugs of SJP has good antibaeterial effect to part of Gram positive bacteria and Gram negative bacteria.Conclusion:SJP has the obvious therapeutic effect on chronic bacterial prostatitis. Its mechanism may be related to its promotion immune, anti-inflammatory effect, increased concentration of Zn2+in the organization.
