| Murdannia divergens (C. B. Clarke) Bruckn is medicine plant from Murdannia genus, there are more than50species in Murdannia genus all over the world,and about18species in our country. Traditional medicinal parts are the dried roots. M. divergens is multi—ethnic drugs commonly used in Yunnan Province. The resources of it are very rich. Many ethnic minorities in the southwest have the histroy of many hundred years on the application of this plant, and the long-term use in all ethnic groups has a good effect. Because of its widely use in the civil, the study on it in gradually increased in recent years.It is very rich in the resource, and its effect is notable, But its use remained in the use of folk, it has not yet seen a detailed study in this thesis, To clarify the character of pharmacognosy and establish an overallã€effective system of quality assessment, ensure the safety and effective in using medicine, provide the scientific basis for exploitation, it was studied systematically in pharmacognosy.we recapitulated the research as the following five parts:Firstly, pharmacognosy Identification:the study on pharmacognosy used character discrimination, micrology discrimination and chemistry discrimination.Second, The herbs conventional check:Determination study of the contents of extracts, moisture, ash of M.divergens referred datas to the Chinese Pharmacopeoia in2010. Result:the average of water-soluble extracts in cold water was18.66%. that in hot water23.75%; the average of eranol—soluble extracts in cold water was9.71%. that in hot water10.09%; the average of ether-soluble extracts was1.08%%, the average of moisture content was11.05%;the average total ashes was15.20%, and the average of acid endurable ashes was6.05%.Third, Determination of total alkaloid content:Use gravimetric analysis to measure the contents of total alkaloid, The different extraction methods were studied for extracting process, The extract of total Alkaloid was determined orthogonal design. The improved process can be described as:me drug powder was refluxed2times,2h each,80%alcohol with19times of the amount to volume of crude drug.Fouth, ITS sequence analysis:The genomic total DNA which are from M. divergens, extracted according to the method of molecular biology. using specific primers for amplifying cloned,purification and sequenced, analysis the sequence of rDNA-ITS.Fifth, Experimental study on antitussive:The cough model induced by ammonia liquor in mice was used to observe the antitussive effects. To compare the relieving cough of different extract isolated from M.divergens, and set up foundation for the experiment which is to search and ascertain the relieving cough.. |
