Objective:To study the best extraction method of total alkaloids of cloud by theorthogonal experiments; to observe the effect of total alkaloids of cloud even onanxiety behavior in mice by the elevated plus maze and clear black box experiment,and to investigate the mechanism of anti anxiety effect by the determination ofneurotransmitters in brain of miceMethod:1.Optimization of the extraction technology of total alkaloids of cloud: To usethe L9(34) orthogonal table, and make the content of the total alkaloid of Coptischinensis as the index of evaluation.To observe the effect of the four factorsconcentration of ethanol (A), ethanol (B), extraction time(C) and extraction times(D) on the extraction process of the total alkaloid of Coptis chinensis Select aoptimum extraction process of the total alkaloid of Coptis chinensis2. Behavior test:Male mice were gastric lavaged with different dosages of thetotal alkaloid of Coptis chinensis for6days.Then use the mouse elevated plusmaze apparatus, light dark box device to observe the cloud even total on the anxietybehavior in mice. Get the whole mouse brain immediately after the behaviorexperiment, and determinate the content of GABA, glutamate and5-serotonin inmouse brain by ELISA.3. To explore the mechanisms of influence of the total alkaloid of Coptischinensis on the anxiety behavior in mice, to analysis the index data by thestatistical method.Result:1.The sequence of factors about the extraction effect of the total alkaloid of coptis chinensis is ethanol concentration> extraction time> ethanol dosage>extraction times. The optimum process of the total alkaloid of coptis chinensis is8times amount of80%ethanol reflux extraction3times, extraction time1h.2. The total alkaloid of coptis chinensis in the elevated plus maze model miceshowed anti anxiety effect, and the effect had no significant difference with thediazepam group. The medium dosage group of the otal alkaloid of coptis chinensiscould significantly increase the content of GABA in the brain of mice, and it had nosignificant difference compared with the positive group. The high dose group andthe medium dosage group of the otal alkaloid of coptis chinensis had the trend toreduce the content of Glu in brain. The three dose groups of the otal alkaloid ofcoptis chinensis had the to reduce the content of5-HT in brain.Conclusion:1.The optimum process of the total alkaloid of coptis chinensis is8timesamount of80%ethanol reflux extraction3times, extraction time1h. The content ofthe total alkaloid in the purified extract reaches above55%.2. The otal alkaloid of coptis chinensis had anti anxiety effect, the mechanismof anti anxiety associated with the increase of GABA content in mice brain, may berelated to the decrease of brain5-HT and Glu content.
