ObjectiveTo investigate the expression and effect of glucagon-like peptide-1receptor(GLP-1R)on sciatic nerve in diabetic rats and the intervention affect of gliclazide.Methods1. Animals Ninty healthy,male Wistar rats weighing160g-180g was used(Supplied byexperimental of animal center of Nan Jing Medical University,Hefei,China)2. Animal Groups Ninty male Wistar rats were modeled with the basic diet feedingadaptation for1week,10specimens from a normal control group (NC group), and theremaining80left modeling.3. Model The rat model of type2diabetes was made by fed with a diet enriched withenergy and ip low-dose streptozotocin.4. Hematoxylin-eosin(HE)staining Histopath-ological changes of sciatic nerve ineach group were observed by lightscope with HE dye.5. Determination of MDA and GSH The application of MDA and GSH kitdetermination of MDA and GSH content, operating in strict accordance with theinstructions.6. Immunohistochemistry Expressions of GLP-1R and caspase-3in each group weredetected by the immunohistochemical method.7. Statistical analysis The data was handled with spss16.0statistic software. Thedifference of many groups was compared with one-factor analysis of variance.The difference of two groups was compared with t-test.The significant level isα=0.05.Theexperimental data were described by means±standard deviation(x±s)Result1. DM rats appear morphological changes of sciatic nerve in the early4weeks by HEdye method, and the damages are aggravated with the extension of DM course. Thechanges in gliclazide intervention group were slighter than model group in thecorresponding period.2. In8weeks model group,MDA content increased significantly and GSH contentdecreased significantly in comparison with the normal control group.In the gliclazideintervention group,the MDA content decreased significantly and GSH contentincreased significantly in comparison with the8weeks model group.3. In8weeks model group,the expression of protein GLP-1R decreased significantly,but the expression of protein Caspase-3increased significantly in comparison with thenormal control group.In the gliclazide intervention groups,the expression of proteinGLP-1R increased significantly but the expression of protein Caspase-3decreasedsignificantly in comparison with8weeks model group.4. When the relative expression amount of GLP-1receptor on diabetic peripheralneuropathy in diabetic rats reduced,MDA content increased, GSH content decreasedand caspase-3protein expression significantly increased.This exception is morepronounced with time.The heavier the rat sciatic nerve lesions,GLP-1R down moreobvious.In the gliclazide intervention group,when the relative expression amount ofGLP-1receptor increased,MDA content decreased, GSH content increased andcaspase-3protein expression significantly decreased.ConclusionIt suggested that GLP-1R expression changes may be associated with DM rats sciaticnerve lesions.GLP-1R can inhibit oxidative stress-induced schwann cell damage and cell apoptosis.It play a protective role in diabetic rat sciatic nerve.Gliclazideintervention can improve the expression of protein GLP-1R,reduced Caspase-3proteinexpression,reduced cell apoptosis,regulated oxidative stress.It have a protective effecton rat sciatic nerve function in DM.
