Objective: Through studying the influence of Yiqihuoxue prescriptionin diabetic rats’ sciatic nerve cell apoptosis and Caspase-3, Bcl-2and Bax,discussing the intervention effect of Yiqihuoxue prescription on diabeticperipheral neuropathy, for the prescription to provide the evidence ofclinical application.Methods:40male healthy Wistar rats, weighting240-260g. Adaptivefeeding one week, according to the random principle to extract10rats andjust as the normal control group, giving the conventional breeding; Theother30rats were made experimental model, feeding high sugar and fatfeed.4th weekend, the experimental group was given the streptozotocinSTZ (dissolved in0.1mol/L citrate buffer, pH4.3,4℃)45mg/kgintraperit-oneal injection, such as the control group only received an equal volume ofcitrate buffer. In the sixth weekend, the experimental rats blood glucosemeasurement was≥16.7mmo1/L, and there is a insulin resistant, namelyinto the model, the successful model rate was90%, a total of27.The27rats were randomly divided equally into three groups, the model group(nine), traditional Chinese medicine group (nine, given Yiqi huoxue prescri-ption25g/kg. d) lavage), the western medicine group (nine, given Methyco-bal0.5mg/kg. d) lavage); the control group and model group rats were giventhe same volume of distilled water,1time/day. Drug intervention for eightweeks. Experimental period, measuring the rat body weight and blood sugar,observing the general condition.The end of the experiment, the level ofneuronal apoptosis was detected by TUNEL method, and immunohistoch-emical method for determining the expression of bcl-2, bax and caspase3.Results:1The general situation of rats During the experiment, the control group rats had normal basic situation;after the first three weeks, the experimental group rats appeared slowly poly-dipsia, diuresis, listlessness, dull matt coat; The fourth week the experimentalgroup by intraperitoneal injection of STZ model four days later, rats appearedunstable, unresponsive, shaggy coat, hip coat color yellow, angular gradually;significant increase in water intake, food intake and urine output, shapelessstool, the cage litter sour symptoms, diabetic peripheral neuropathy sympto-ms appeared. To start from the sixth week to give the drug intervenetion for aperiod of eight weeks, in addition to the model group, traditional Chinesemedicine group and western medicine group symptoms of rats graduallyreduced, the effect is more obvious with prolonged treatment.2The changes of body weight, blood sugar in rats2.1The changes of body weight in rats1weekend, each group of the body weight difference was not statistical-ly significant (P>0.05).4weekends, the body weight difference was statisti-cally significant (P <0.001). Compared with the normal control group, the bloodsuger in each group rats was increased (P <0.05); Among the model group,the western medicine group and the Chinese medicine group, the differencewas not statistically significant (P>0.05).6weekend, the blood sugardifference was statistically significant (P <0.001). Compared with the normalcontrol group, the blood suger in each group rats was increased (P <0.05);Among the model group, the western medicine group and the Chinesemedicine group, the difference was not statistically significant(P>0.05). Drugintervention for eight weeks, difference was significant between groups,statistical significance (P <0.001). Compared with the model group, thetraditional Chinese medicine group blood suger was significantly lower (P <0.05); the western medicine group to lower blood sugar meaningful, but notobvious (P>0.05); The Chinese medicine group had better hypoglycemiceffect to western medicine group (P <0.05).3The detection results of sciatic nerve apoptosisThe normal control group sciatic nerve apoptosis was less, the apoptosis ineach group rats was significantly increased (P <0.05); Compared with themodel group, Methycobal group and Yiqihuoxue prescription group ratsapoptotic cells were significantly reduced (P <0.05), the traditional Chinesemedicine group was more apparent.4The change of Caspase-3, Bcl-2and BaxCompared with the normal control group, the remaining three groups ofCaspase-3, Bax expression were significantly increased, Bcl-2significantlydecreased, there was a statistically significant difference (P <0.05). Comparedwith the model group, the Chinese medicine group and western medicinegroup of Caspase-3,Bax expression was significantly decreased, and Bcl-2was obviously improved, the difference was statistically significance; thetraditional Chinese medicine group had more significant difference to theMethycobal group.Conclution:1To the general situation of diabetic rats, the body quality, blood suger,Yiqi huoxue prescription intervention role better than Methycobal.2The sciatic nerves exist a wide range of apoptosis, resulting in DPN.3Cell apoptosis is regulated by Caspase-3, Bcl-2and Bax.4Yiqihuoxue prescription can improve the diabetes rats sciatic nerveapoptosis and the expression of regulation factor Caspase-3, Bcl-2and Bax.A certain role in the prevention and treatment of DPN.
