Streptococcus mutans is a cariogenic bacteria in the human oral cavity,by detecting thenumber of t hem we can evaluate people’s caries activity and predicting risk ofindividuals dental caries. Thus it is able to provide an important reference for themonitoring and prevention of dental caries. Microfluidic chip technology allows theentire biochemical reaction is concentrated in a micron-scale chip. Due to it’scharacteristics of rapid, sensitive, low cost and automated analysis, has developed intoone of the popular directions of multidisciplinary research.The issue group had used fluidic electrode microfluidic chip based on electrochemicalmethod for rapid quantitative detection of the main pathogens of chronic periodontitis-Porphyromonas gingivalis, but whether this method applies to the detection ofStreptococcus mutans is not clear. So we introduce electrochemical method into thisfield by our research. In experiment one, we use fluidic electrode microfluidic chip as aplatform to complete the quantitative detection of Streptococcus mutans and then compare the result to bacterial culture for verifying the accuracy and applicability ofthis method.Currently the most widely used in the microfluidic chip detection is mainly metalelectrodes, including gold electrode, platinum electrode, copper electrode, nickelelectrode, aluminum electrode and ITO electrode. Gold and platinum due to it’s stablechemical properties, easy processing and low noise advantages, is the most widely used,but the price is more expensive. ITO (In2O3: SnO2=9:1) film with high conductivity,high visible light transmittance, high infrared reflectivity, combine with glasssubstrate strong, scratch resistance, including many other excellent physical propertiesand good chemical stability as well as some other semiconductor properties, easypreparation into the electrode pattern and the economic applicability, concurrently. Inexperiment two, we use ITO solid metal electrode microfluidic chip as a platform toquantitative detection of Streptococcus mutans to verify the accuracy and applicabilityof electrochemical method once again.The results of this study show that based on microfluidic chip with electrochemicalmethod for bacterial detection can distinguish104-109cells/ml concentration ofStreptococcus mutans. We obtain stable values of the detection time about ten minutes.They have a highly positive correlation between two kinds of method. Electrochemical method based on microfluidic chip for bacterial detection applied tothe field of quantitative detection of Streptococcus mutans for the first time by writer.Not only providing new ideas for researchers in this field, but also obtaining a certainexpansion with the scope of application by this method. Thus provide a effectivereference for the further application of the method in the field of oral bacteria detection.
