ObjectiveTo study the effects of PSK on blood lipids and hemorheology in therats with hyperlipidemiaMethodsForty male SD rats with weight of80±20g were used. The bloodsampleswere taken from the tails of rats that were starved for12h afterthey were fed with ordinary food for one week and blood lipids weredetermined. The rats then, were randomly divided into5groupsaccording to the blood lipid values:normal diet group, high fat andcholesterol diet group, high fat and cholesterol diet plus lovastatin7.2mg/kg group, and high fat and cholesterol diet plus PSK groups with either500mg/kg or2500mg/kg. All the drugs were prepared as suspensionsolutions with0.5%sodium carboxymethylcellulose (CMC). The high fatand cholesterol diet contained1.5%cholesterol,0.25%cholalic acids,5%sugar,5%lard,5%eggs, and0.2%propylthiouracil. At the90thd,120thd,and150thd after drugs were given, the TC, TG, LDL-C, and HDL-C fromrat blood samples were determined with determinqation ofhemorheological parameters at the150thd.Results1. The high fat and cholesterol diet used made the fed rats elevationof serum TC and LDL-C significantly in two months, and the values of TCand LDL-C were5.2times and8times than that of rats with normal diet5 months later. However, the high fat and cholesterol diet made rat serumTG decreased in time-dependent manner.2. PSK inhibited elevation of serum lipids caused by high fat andcholesterol diet in dose-and time-dependent manners, and showedsignificantly decreasing blood lipids5months after it was given. Besides,two doses of PSK markedly raised levels of HDL-C and prevented adiposedegeneration of liver caused by high fat and cholesterol diet.3. There existed clearly increasing whole blood viscosity, bloodplasma viscosity, hemotocrit, erythrocyte aggregation index, anderythrocyte electrophoretic time in hyperlipidemic rats. Pretreatment ofPSK with either500mg/kg or2500mg/kg significantly blunted theelevation of hemorheological parameters above.Conclusion1. A successful rat hyperlipidemic model is set up, which is suitable toinvestigate the drugs that have modulation of serum TC, LDL-C, andHDL-C.2. Pretreatment of PSK has a good modulation of blood lipids andsignificantly decreases TC, LDL-C with increasing HDL-C.3. Pretreatment of PSK also modifies the hemorheological parametersand decreases viscosity, coagulation, and aggregation of blood.
