We investigated the effects of Ganoderma tsugae polysaccharide on hyperlipidemia andatherosclerosis in mouse and rats model, and compared the results of different administration(ig or ip) and different dose groups. Through different administration with oral intubation andintraperitoneal paracentesis and different dose compare with inter groups. Simvastatin wasused as positive control. The concentrations of total cholesterol, triglyceride, low densitylipoprotein cholesterol and high density lipoprotein cholesterol were investigated bybiochemical methods. The effect of Ganoderma tsugae polysaccharide on the blood lipid wasstudied, and the primary mechanism was also discussed. The effect on antiatherosis wasobserved by hemetoxylin and eosin stain, immunohistochemical method and opticalmicroscope observation.The results showed that the Ganoderma tsugae polysaccharides can decrease the levels ofTC, TG and LDL-C in serum, and enhance the level of HDL-C. The pathological results ofthe polysaccharide groups showed the three-layer construction of blood vessel wall was clearand complete, thickness of plaque was thinner and protuberant were limitation; the foam cellwas few, the lipid storage was rare and the smooth muscle cells lined up out of order.The statistical results showed that the effects of regulated-blood lipid and anti-atherosisare obvious, whether in SSFA,SSG oral intubation group or in SSF intraperitonealparacentesis group. Compare with simvastatin group, there was no obvious difference.According to TC, the effects of SSFA, SSG 25mg/kg,50 mg/kg and SSF 5mg/kg dose groupwere more excellently. The expressions of HMG-CoA and FAS mRNA were decreased indifferent degree by RT-PCR. The expression of ApoA protein was increased and theexpression of ApoB protein was immovability by biochemical assay.Conclusion: Ganoderma tasugae polysaccharides can reduced the levels of LDL,TC,TG in serum in rats and mouse model of hyperlipidemia, it is an effective compound forregulating blood lipid and antiatherosclerosis. Our data indicated the mechanism thatGanoderma tsugae polysaccharides regulated the blood lipid and antiatherosclerosis isreducing the synthesis of TC and increasing the expression of ApoA protein. But themechanism of polysaccharide decreased the serum LDL-C is still not clear. The results ofpathology or the pathological results showed that Ganoderma tsugae polysaccharide is of notoxic and adverse effect to heart, liver, spleen, lung, kidney in rats.
