A Study On The Relationship Between The Expression Of MIF, MMP-9, VEGF And Helicobacter Pylori L-form Infection In Gastric Carcinoma

Objective:Study the expression of MIF, MMP-9, VEGF in the gastric carcinoma and clinical significance, and explore the relationship between the MIF, MMP-9, VEGF expression and Helicobacter pylori L-type infection in gastric cancer.Methods:(1) Collected80cases of paraffin blocks of gastric cancer surgical specimens and associated clinical and pathological data, and randomly selected50cases of peripheral tissues as a control group, detected the protein in the organizations of MIF, MMP-9and VEGF by immunohistochemistry (Elivision France).(2) Collected30cases of fresh gastric cancer tissues and corresponding peripheral tissues (normal control), and using reverse transcription polymerase chain reaction, detected the MIF-mRNA expression in these organizations.(3) Gram staining and immunohistochemical assay of the80cases of paraffin organization of Hp-L-type infection.Results:(1) The results of immunohistochemistry showed that:the gastric cancer group, MIF, MMP-9, VEGF positive expression rates higher than the control group (P<0.05); and the expression of the three with the depth of tumor invasion, lymph node metastasis (P<0.05), which MIF-positive expression rate in addition to the depth of tumor invasion, lymph node metastasis the outside with the degree of tumor differentiation, clinical stage (P<0.05), has nothing to do with gender, age (P>.05), MMP-9expression level and depth of tumor invasion, lymph node metastasis, clinical stage (P<0.05), has nothing to do with gender, age, tumor differentiation degree (P>0.05), and VEGF expression levels only with the depth of tumor invasion, lymph node metastasis (P<0.05), and gender, age, tumor differentiation, clinical stage unrelated (P>0.05).(2) The results of RT-PCR results showed that:the gastric cancer group, MIF mRNA expression level higher than that in the control group (P<0.05), and the value of MIF mRNA and depth of tumor invasion, lymph node metastasis (P<0.05), gender, age, tumor differentiation, clinical stage etc. unrelated (P>0.05).(3) Hp-L test results show that:Hp-L-type results of Gram staining, immunohistochemistry, two methods (P<0.05), Hp-L infection positive rate in gastric cancer group and control group respectively71.25%,14.00%, both compared with the statistically significant (P<0.05), and gastric cancer group Hp-L expression only with lymph node metastasis (P<0.05).(4) Gastric cancer group of Hp-L-type infection-positive by the MIF, MMP-9, VEGF positive rate was84.20%,96.5%,94.7%, higher than the L-type Hp-negative (positive rate21.70%,78.3%,78.3%,P<0.05), and Hp-L type infection and the MIF, MMP-9, VEGF positive expression was positively correlated (r=0.598, r=0.292, r=0.249,P<0.05).(5) Gastric cancer patients MIF positive MMP-9, VEGF positive rate was96.2%,98.1%, higher than the MIF negative (positive rates were81.5%,74.1%,P<0.05), gastric cancer MIF and MMP-9, VEGF expression was positively correlated (r=0.340, r=0.379,P<0.05).Conclusion:1. MIF, MMP-9and VEGF in gastric cancer tissues are highly expressed, and all with the depth of invasion, lymph node metastasis, suggesting that the three may play a role in promoting invasion and metastasis of gastric cancer.2. The expression of MIF and MMP-9、VEGF in gastric cancer were positively related, suggesting that MIF may be through up regulation the expression of MMP-9, VEGF in promoting tumor invasion, lymph node metastasis.3. Hp-L infection rate of gastric cancer group was significantly higher than that associated with lymph node metastasis, indicating that the Hp-L infection in gastric transfer process.4. Gastric cancer group Hp-L infection positive rate and the MIF, MMP-9, VEGF expression showed a positive correlation, suggesting that Hp-L infection to promote tumor invasion, the transfer mechanism and increase the MIF, MMP-9, VEGF expression.
