ObjectivesStudy of Simotang oral liquid on isolated rat jejunal smooth muscle motion effects, to explore its mechanism, which helps to reveal the effect of Simotang modulates gastrointestinal motor pathways and mechanisms, as in a development Chinese traditional medicine in the treatment of functional gastrointestinal disorders with modern scientific basis and theoretical support.MethodsUsing the hanging method, observation of Simotang oral liquid after the administration of the drug in vitro smooth muscle contractile function, observed in different Ca2+、K+environment and α、β、M receptor antagonist, agonist case of Simo decoction on rat jejunum smooth muscle contraction motion effect.Results1.Simo decoction on isolated rat jejunal smooth muscle contraction movement of low dose, high dose suppression promote two-way adjusting effects.2. Acetylcholine can make the spontaneous contraction of stable rat intestinal isolated smooth muscle spasm, delivery and contraction amplitude and frequency were visible very significant difference(P<0.01); Simo decoction can inhibit the spasm, delivery and contraction amplitude and frequency were visible very significant difference(P<0.01).3. Effect of atropine on spontaneous contraction of stable rat intestinal isolated smooth muscle exercise has significant inhibiting effect, after the administration of the drug contraction amplitude comparison shows significant differences(P<0.01); Simo decoction on atropine after inhibition of smooth muscle exercise effect is not obvious, contraction amplitude comparison before and after drug administration had no significant difference(P>0.05).4. Adrenergic inhibition of spontaneous contractions of stable rat intestinal isolated smooth muscle movement, delivery and contraction amplitude comparison shows significant differences(P<0.01); Simo decoction on adrenergic inhibition of smooth muscle has stimulative effect, after the administration of the drug contraction amplitude comparison see very significant difference(P<0.01).5. Phentolamine on spontaneous contraction stability in rat intestinal smooth muscle motility in vitro contraction amplitude and contraction frequency after the administration of the drug compared had not obvious change, no statistical significance(P>0.05); Simo decoction on phentolamine after muscle movement has the effect of promoting the role, after the contraction amplitude and the previous administration results show significant differences(P<0.01); Simo decoction after administration with spontaneous contraction amplitude comparison also has a very significant difference(P<0.01); the contraction frequency of two groups before and after drug administration comparison had not obvious change, no statistical significance(P>0.05).6. Calcium chloride on the spontaneous contractile stability in rat intestinal smooth muscle motility in vitro contraction amplitude and frequency of contraction after the administration of the drug compared had not obvious change, no statistical significance(P>0.05); calcium chloride effect of Simotang on the smooth muscle contraction amplitude and dosing before more visible very significant difference(P<0.01), Simo decoction after administration with spontaneous contraction amplitude comparison shows significant differences(P<0.01); the contraction frequency of two groups before and after drug administration had no statistical significance(P>0.05).7. Rat intestinal smooth muscle in vitro calcium-free Tyrode’s solution of motion is not obvious, the addition of calcium chloride on its motion effects are not obvious, no statistical significance(P>0.05); on the basis of this added Simotang the amplitude of contraction and dosing before more visible very significant difference(P<0.01).8. Potassium chloride can inhibit the spontaneous contraction of stable rat intestinal isolated smooth muscle movement, make the contraction amplitude was significantly reduced after the administration of the drug, the contraction amplitude comparison of visible very significant difference(P<0.01); Simo decoction on the potassium chloride after inhibition of smooth muscle has stimulative effect, after the administration of the drug contraction amplitude comparison see significant difference(P<0.01); the contraction frequency of two groups before and after drug administration had no statistical significance(P>0.05).9. Verapamil can complete inhibition of spontaneous contractions of stable rat intestinal isolated smooth muscle movement, the amplitude of contraction amplitude becomes zero, delivery and contraction amplitude comparison shows significant differences(P<0.01). Simo decoction on verapamil inhibits smooth muscle movement had no effect after.Conclusion1.Simo decoction on isolated rat jejunal smooth muscle contraction movement of low dose, high dose suppression promote two-way adjusting effects.2.Simo decoction on isolated smooth muscle of rat intestinal facilitation effect might be mainly with M cholinergic receptors, adrenergic receptor, may be mediated through Ca2+channels to open, to promote Ca+influx and/or Ca2+release and promote intestinal smooth muscle.
