Objective To study the effect of continuous regional arterial infusion (CRAI) of dexamethasone on plasma Inflammatory factors of rabbits with severe acute pancreatitis (SAP) rabbits.Methods Twenty-four new Zealand white rabbits (2.5-3.5kg) were provided and feed by GLP laboratory of medical department of lanzhou university. There were half of male and female. They were forbidden to eat but drink12hours before experiment.They were randomly divided into sham operation group(SO group). SAP group, intravenous infusion of dexamethasone group(IVI group) and CRAI of dexamethasone group (CRAI group)(each group6rabbits) according to random number table. The serum tumor necrosis factor (TNF)-a, interleukin (IL)-6, IL-10levels in rabbits were tested at hour0.5,3,6,9and12h after modeling succeed.The pathological changes of pancreas were observed under Light microscopy.Results Compared with SO group, SAP group:the serum levels ofI1L-6of significantly increased at3h and reached the peak at6h,decreased at9h(all P<0.05); the serum levels of IL-10significantly increased at6h, continuously elevated at9h and12h (all P<0.001); the serum levels of TNF-a significantly increased at0.5h(P<0.001),reached the peak at6h(P<0.001) and decreased at9h(P<0.05) Compared with the SAP group, CRAI group:the serum levels of IL-6and IL-10all significantly decreased at6h,9h,and12h(P<0.001);IVI group:the serum levels of IL-6significantly decreased only at6h(P<0.05), the serum levels of IL-10were not changed all the time (P>0.05); the serum levels of TNF-a in the CRAI group significantly decreased at3h,6h,9h.and12h(all P<0.001), which in the IVI group significantly decreased only at6h (P<0.05).Compared with IVI group, the CRAI group:the serun levels of IL-6and IL-10all significantly decreased at6h (P<0.05)and12h (P<0.001);levels of TNF-a all significantly decreased at3h,6h,9h,and12h(all P<0.001). The pathological changes of pancreas in the CRAI of dexamethasone group were obvious.Conclusion Continuous regional arterial infusion dexamethasone can effectively reduce the systemic inflammatory response and pancreatic inflammation.
