Objective:To observe the expression of mutational PBRM1and mutational P53in different stages and classification of renal cell carcinoma (RCC) and normal tissue,to discuss the expression of PBRM1and P53in RCC and their correlation.Methods:Expression of PBRM1and P53was detected by immunohistochemical technique in66cases of RCC and20cases of normal renal tissues,Selected24cases fresh renal clear cell carcinoma samples and10cases of normal fresh renal tissue samples were detected by retrovirus-polymerase chain reaction (RT-PCR) technique.Detected gene expression quantity of PBRM1mRNA and P53mRNA in these tissues.Analysis the relationships and correlation of the two index and RCC clinical pathology.Results:1.The expression of PBRM1and P53protein in different pathological changes renal tissuesP53and PBRM1protein expression were62.1%and39.4%in the RCC tissues. PBRM1expression was associated with RCC grade and stage (P<0.05),was not associated with RCC pathological types(P>0.05).P53expression was irrelevant to RCC grade and stage(P>0.05)2.The expression of PBRM1mRNA and P53mRNA in different degree pathological changes renal tissues.Used light density to analysis the channel of agarose gel electrophoresis,the results showed that the expression of PBRM1mRNA and p53mRNA in RCC tissues was significantly higher than normal renal tissues.3.Analysis the correlation of PBRM1and P53proteinPBRM1and p53combined expression in RCC was showed in table3, both the same expression rate was70.83%(17/24). If the weak positive and strong positive calculated for common, both the same expression rate was83.33%(20/24). Spearman correlation analysis showed that the expression of PBRM1and P53in RCC was positively correlated (r=0.770, P<0.001)Conclusion: The expression level of PBRM1was increased with RCC tissues pathology classification and the clinical stages,may be related with the biological action of the tumor.And the expression of P53in RCC tissues was higher than normal renal tissues,but it was not important meaning in the RCC biology behavior.
