Objective:To study on the expression and clinic significance of extracellular matrix metalloproteinase inducer(EMMPRIN) in renal clear cell carcinoma(RCCC) and paracarcinomatous renal tissue(PCRT),to investigate the possible molecular mechanism of EMMPRIN in the RCC growth and recurrence/metastasis,we are aiming at providing a theoretical basis to the molecular target for the treatment of renal cell carcinoma.Methods:Forty-three RCC and Ten PCRT were obtained underwent radical nephrectomy for renal carcinoma and confirmed to be RCCC between 2008.4~2009.4 in the Union Hospital of Huazhong University of Science and Technology. Reverse transcription polymerase chain reaction and immunohistochemistry was performed for all the samples so as to explore the EMMPRIN mRNA and protein expression level, and to analyze the correlation of the EMMPRIN mRNA and protein expression level with clinlic pathology. Results:Expression of the EMMPRIN mRNA and protein were noted in both RCCC and PCRT: The positive rates of EMMPRIN mRNA in RCCC was 60.0%,much higner than in PCRT(20.0%,P<0.05).The positive rates of EMMPRIN protein in RCCC was 55.8%,much higner than in PCRT(20.0%,P<0.05). The expression of the EMMPRIN mRNA and protein was merely correlated with the clinlic grades and pathology stages(P<0.05) in RCCC,but not with the age, gender and tumor site(P>0.05).Conclusion:The study demonstrated the high expression of the EMMPRIN mRNA and protein, and both expression levels were correlate with the clinic grades and pathology stages significantly, but not with the age, gender and tumor site. This research indicated that EMMPRIN can be a marker in the RCC ocurrence, growth and metastasis, and may be a potential target for the treatment of RCC.
