Research background:Renal cell carcinoma(RCC)is a common malignant tumor with a high incidence in the world.Studies have shown that PBRM1 is a gene with a high mutation rate in patients with RCC.Currently,most studies on PBRM1 are based on the transfer of a small number of clinical samples to evaluate its influence on patients,and few clinical studies based on multiple copies and big data in databases are used to evaluate the influence of differential expression of PBRM1 in RCC patients on the prognosis and survival of patients.Objective:The aim of this study was to analyze whether the differential expression of PBRM1 gene affected The prognosis and survival length of patients with renal cell carcinoma(RCC)based on National Cancer Institute(NCI)surveillance data and The Cancer Genome Atlas(TCGA)data.The influence of this gene on patients with renal cell carcinoma was comprehensively evaluated after grouping according to different major subtypes of renal cell carcinoma,gender,tumor progression,etc.The results were compared with genetic data and immunohistochemical data from The Human Protein Atlas(HPA)database.Methods:A total of 890 cases of renal cell carcinoma(RCC)from 2006 to 2020 were included in the TCGA database,and classified according to different subtypes.There were 533 cases of renal clear cell carcinoma(KIRC),290 cases of Kidney papillary cell carcinoma(KIRP),and 67 cases of Kidney chromophobe(KICH).By matching the target gene expression data of normal kidney tissue,the expression difference between each subtype of cancer tissue and normal kidney tissue was compared.Then,they were grouped according to different factors such as gender,tumor grade,tumor progression,etc.,the results were compared and the differences were verified,and the survival curves of patients with renal cell carcinoma under different expression levels were drawn and tested.In terms of verification of relevant statistical data and results,the target gene data in the HPA database were used to draw interactive survival scatter plots and survival curves under the influence of differential expression of this gene.At the same time,immunohistochemical samples of some cases of renal cell carcinoma selected from the database were selected to observe the number and intensity of antibody staining in the statistical samples,which were compared and verified with the statistical analysis of TCGA data.Results:The expression level of PBRM1 gene in different types of renal cell carcinoma was lower than that in normal renal tissue,and the difference of expression level was statistically significant.This result reflected from the side that PBRM1 gene can inhibit the occurrence or development of cancer in patients with renal cell carcinoma.The absence or mutation of the gene lead to the lower expression of may promote the production and the progress of tumor.The expression level of PBRM1 gene in renal clear cell carcinoma was significantly lower than that in normal renal tissues and other subtypes of carcinoma tissues,which also indicated that PBRM1 gene may have a higher mutation rate in renal clear cell carcinoma.After the survival curve was drawn and tested,it was found that the differential expression of PBRM1 gene had an effect on the survival time of patients with renal clear cell carcinoma(P=0.031),that is,the high expression of PBRM1 gene had a positive effect on the prolongation of the survival time of patients with renal clear cell carcinoma.In papillary renal cell carcinoma and renal chromophobe cell carcinoma,however,differential expression of this gene does not actually affect survival.Conclusion : 1.PBRM1 gene plays an inhibitory role in the development and progression of renal cell carcinoma.2.The expression level of PBRM1 gene in tumor tissues of patients with renal cell carcinoma was lower than that in normal kidney tissues,indicating differential expression,and the difference was statistically significant(P<0.05).3.Patients with high PBRM1 gene expression in renal clear cell carcinoma have better prognosis and longer survival.
