Objective To investigate the expressions of peripheral blood mononuclear cells(PBMCs) interleukin-23p19mRNA and plasma interleukin-23in patients with type2diabetes mellitus(T2DM) before and after therapy, further explore the relationship between interleukin-23and T2DM during the occurrence and development.Methods The study applied retrospective case-control study,the case group selected58patients with T2DM, the fasting plasma glucose level in selected grouping objects,28cases with patients of untreated T2DM are frist medical of the outpatients and in hospital patients,the fasting plasma glucose level3.9-6.1mmol/1.30cases with already treated for T2DM and the fasting plasma glucose level=7.0mmol/1are already treated for T2DM.26cases of healthy check-up for the control group.We collected the EDTA-K2peripheral blood,extracted total RNA from PeriPheral blood mononuclear cells. Levels of interleukin-23p19mRN A were Measured by reverse transcription-Polymerase Chain Reaction (RT-PCR). The plasma of interleukin-23levels were typed using enzyme-linked immunosorbent assay(ELISA).Results①According to fasting plasma glucose level group selected three group of subjects, the levels of interleukin-23mRNA were significantly increased in patients of untreated T2DM compared with after treatment and control(p<0.05).②The expression levels of interleukin-23mRNA were no significant difference in treatment group and control(P>0.05).③the plasma levels of interleukin-23in newly diagnosed T2DM were significantly higher than those in after treatment and control (P<0.05).④The plasma of interleukin-23levels were found no significant difference between the treatment group and control group.Conclusions The levels of interleukin-23and p19mRNA were significantly increased in patients of untreated T2DM compared with after treatment and control,IL-23may play an important role in T2DM patients did not use drugs and treatmenet, IL-23may be involved during the occurrence and development of T2DM.The level of fasting blood glucose in untreated T2DM beyond the normal range,however,the group of after treatment control in the normal range and the control group is normal,that IL-23may exist some Relationship with fasting blood glucose.
