Background:There are various causes of tissue’s low blood perfusion that lead to ischemic injury reduction cell, the focused point to solve the ischemic injury is the early restoration of blood supply,but when ischemia reperfusion after restoration of the blood Cell ultrastructure, functional, metabolic and electrophysiological aspects will further be damaged and aggravated,this injury phenomenon is called ischemia reperfusion injury. Severe limb ischemia reperfusion injury can cause lung, kidney, heart, pancreas, spinal cord and distant organs and tissues damage, what is more,result in systemic inflammatory response syndrome, and multiple organ dysfunction syndrome.Therefore research on ischemia-reperfusion injury prevention is very important.Because of its side effects and other reasons, western medicine for the prevention and treatment of ischemia reperfusion injury have not yet achieved a better clinical results right now.Because of its fewer side effects, a variety of traditional Chinese medicine on the treatment of limb ischemia reperfusion injury has unique advantages.Clinical trials have tanshinone ⅡA sodium sulfonate injection in cardiovascular disease prevention and treatment, and obtained the certain effect.But at present,domestic and international of tanshinone ⅡA trials on ischemia reperfusion injury and protective effect focused mainly on the brain, but the lack of limb ischemia reperfusion injury. It is significanct to study the tanshinone ⅡA sodium sulfonate injection on limb ischemia reperfusion injury and the protective effect and its mechanism.Objective:To study the effect and the mechanism of Sodium Tanshinone ⅡA Silate on limbs ischemia-reperfusion injury of rats.Methods:The48healthy Wistar male rats were randomly divided into three groups (n=16): the tanshinone group, the control group and the sham group. In the preoperative3,2,1day and preoperative30minutes, the tanshinone group was given intraperitoneal injection of Sodium Tanshinone ⅡA Silate injection (20mg/kg); the control group and the sham group at the same time given intraperitoneal injection of normal saline. The tanshinone group and the control group to establish limb ischemia-reperfusion injury model, at2hours post-ischemia and4hours post-reperfusion sample of blood serum was collected for the measurement of SOD activity and the contents of MDA through the chemical methods. Meanwhile the skeletal tissue was used for the light microscope examination.Results:Compared with the sham group, SOD activity decreased and the contents of MDA increased on different phases in the control group. Compared with the control group, SOD activity increased and the contents of MDA decreased on different phases in the Sodium Tanshinone group. SOD activity was obviously decreased while the contents of MDA were obviously increased in the4hours post-reperfusion compared with the2hours post-ischemia in the control group. SOD activity and the contents of MDA showed no significant change in the4hours post-reperfusion and the2hours post-ischemia in the tanshinone group. Under light microscope sham operation group rat hindlimb skeletal muscles even dyeing, muscle fibers are arranged in rows, striated muscle cell nucleus clearly visible.the control group of muscle fiber edema, most striated disappeared, cytoplasm pale, part of the muscle loss of nucleus;the tanshinone group of muscle fiber edema significantly reduced, most of striated muscle more clearly visible.Conclusion:Sodium Tanshinone ⅡA Silate injection can make the rats serum SOD activity increased, MDA decreased, loss of skeletal muscle tissue damage, and has protective effects in rat limb ischemia reperfusion injury.
