Coronary heart disease is mainly caused by the atherosclerosis. Atherosclerosis, namely adeposition of a layer of millet-gruel-like lipid under the artery wall, can reduce the elasticityand calibre of an artery. Large and medium muscular elastic arteries such as aorta, coronaryartery and cerebral artery are easier to be subjected to AS, resulting in some severeconsequences, for example occlusion and rupture of blood vessels. AS is often discovered inmen over forties or postmenopausal women. It is usually coupled with hypertension,hyperlipidemia, diabetes and so forth. Unfortunately, AS has become one of main deathreasons among old people and patients in western countries. With the improvement of livingstandards and changing diets of Chinese people, AS has also got its steady foothold amongpatients in China. Investigating into its etiology, we discover that AS involves theparticipation of various cells and molecules. According to a recent research, PI3K is asignificant part of the intracellular signal transduction pathway PI3K-Akt which participatesin many physiological and pathological processes such as proliferation, apoptosis, migration,inflammation and so on. Thus have a tight connection with development of AS. Therefore,PIK3CG gene polymorphisms probably relates to AS disease.Objective: To investigate the association between PIK3CG gene polymorphisms andcoronary heart disease, and the association between coronary heart disease and lipidmetabolism. To analyze the association between coronary heart disease and environmentalfactors, and association of PIK3CG gene polymorphisms and lipid metabolism clinicalindexes acquired from coronary heart disease patients; analysis of association of PIK3CGgene polymorphisms and environmental risk factors.Methods: In this study, we chose northern people group in China as objects. PIK3CGgene and four tag SNPs rs1526083, rs3173908, rs6961244and rs12667819on the gene arethen selected to analyse the association between PIK3CG gene polymorphisms and coronaryheart disease by case-control study. This method is also applied to studies of associationbetween coronary heart disease and lipid metabolism clinical indexes and relatedenvironmental factors. Case-only study is used in analysis of association of PIK3CG gene polymorphisms with lipid metabolism clinical indexes and its association with relatedenvironmental factors.Results: In this study, except for rs3173908, the frequence distribution of genotypes andalleles of3positions rs1526083, rs6961244and rs12667819all show an accordance withH-W balance in both control and case groups(P>0.05). Association analysis hasdemonstrated that there is no statistical significance in the frequence distributionaldifferences of the genotypes and alleles in control and case group (P>0.05). Multi-lociassociation analysis indicates there is no association between the6haplotypes on PIK3CGand coronary heart disease (P>0.05). We can also find that no association exists betweenPIK3CG gene polymorphisms and blood lipid level of coronary heart disease patients bycase-only study. Further more, smoking and drinking of patients doesn’t interact withPIK3CG gene polymorphisms (P>0.05). An obvious association between coronary heartdisease and abnormal TC, TG, HDL-C level and its related environmental factor smokingcan be discovered by case-control study(P<0.05).Conclusion: PIK3CG is possibly not a susceptible gene of coronary heart disease.However, it is not excluded that PIK3CG may be one minor gene of plenty of susceptiblegenes of coronary heart disease. Because coronary heart disease is a result of synergy of arecord number of minor genes, thus one single gene can have weak influence on it.Meanwhile, differences resulting from races and religions exist in various susceptible genesof coronary heart disease. In this study we choose only northern people of China as objects,so PIK3CG polymorphisms of other people groups may have association with coronary heartdisease. There is an obvious association between coronary heart disease and TC, TG, HDL-Clevel and smoking. PIK3CG cannot influence blood lipid level of northern people in China,therefore it cannot contribute to AS by changing blood lipid level. PIK3CG polymorphismsmay not interact with smoking or drinking to influence the development of coronary heartdisease.
