Purpose: To research association between AS and PTEN gene polymorphism, andassociation between PTEN with metabolic index and coronary heart disease among patients.To analyze of interactions of PTEN and environmental risk factors.Methods: In this study people of China’s main ethnic group in Beijing are regarded asthe objects, we choose two tag SNPs rs532678and rs1234219on PTEN to analyze relationsbetween PTEN polymorphism and CHD by case-control methods. Discuss associationbetween PTEN polymorphism and coronary heart disease according to frequencydistribution of the genotype which contains two different positions among coronary heartdisease patients. Analyze interactions of PTEN and environmental risk factors via case onlystudy. Provide important clues to studies on etiology and basis of prevention and therapy ofcoronary heart disease.Results: In the study, the frequency distributions of the genotype and its alleles in bothcase group and control group are all corresponded to H-W balance(P>0.05). Throughassociation analysis it can be found that there are no statistical significances in frequencydistributional differences of the genotypes of different positions and their alleles in both caseand control group(P>0.05). There are also no statistical significances in distributionaldifferences of one haplotype system consisted of two positions on PTEN in bothgroups(P>0.05). Male rs532678position polymorphism does not possibly have connectionswith coronary heart disease; female rs532678polymorphism is perhaps associated withcoronary heart disease; rs1234219polymorphism is likely not related to coronary heartdisease. Among blood lipid indexes, TC and TG level is related to coronary heart disease,however, LDL-C and HDL-C does not bear on the disease. Analysis of interactions of PTENand environmental factors by way of case only study indicates that a positive interactionexists between rs1234219polymorphism and female smoking. There is a synergistic actionbetween the two factors but interactions of its polymorphism and coronary heart disease’senvironmental risk factors are not found. We can discover that PTEN polymorphism may beindependent from TC, TG and HDL-C indexes mutually; nevertheless there is a synergisticaction among rs532678position’s TC+CC genotype and LDL-C index and rs1234219polymorphism is possibly independent from LDL-C index, by analysis of relationship between the gene and blood lipid indexes.Conclusion: From this study, we conclude that there is association between PTEN genepolymorphism and coronary heart disease and it may be a susceptible gene of coronary heartdisease. Female rs532678polymorphism may has connections with pathogenesis of coronaryheart disease. Female TC, TG and HDL-C level is related to pathogenesis of the disease. Apositive correlation exists between smoking and morbidity rate of coronary heart disease.There is a synergistic action among rs1234219polymorphism on PTEN and female smoking.And it can be also concluded that a synergistic action exists between TC+CC genotype ofrs532678and LDL-C level.
