| Objective:To observe the intervention of atorvastatin and CoQ10on myocardial energymetabolism in rabbits with hypercholesterolemiaMethods:fourty male New Zealand white rabbits were randomly divided into fivegroup:control group (group A),cholesterol group (group B),cholesterol+atorvastatintreated group (group C),cholesterol+atorvastatin+average dose CoQ10group(groupD),cholesterol+atorvastatin+high dose CoQ10group(group E).After feeding for6weeks,the empty stomach blood preparation through ear marginal vein were collected,thelevels of serum total cholesterd were determined;the myocardium was taken to observedits ultrastructures by electron microscopy;High performance liquid chromatography(HPLC) was used to measure myocardial mitochondria ATP and CoQ10;Ultravioletspectrophotomtry was used to measure the activities of SDH ang CCO.Results:(1) in group B,myocardial fibers derangement,part of thefracture,dissolution,mitochondria swelling,crest disturbance,fuzzy,compared with groupA,the activities of SDH and CCO were significantly decreased,the content of myocardialmitochondria ATP and CoQ10was ignificantly decreased.(2)compared with group B, the activities of SDH and CCO in group C wereincreased, compare to the content of myocardial mitochondria ATP and CoQ10,there wasno significant difference between the two groups.(3)Compared with group C,the content of myocardial mitochondria ATP andCoQ10in group D E were increased,compare to the activities of SDH and CCOthere was no significant difference between the two groups.(4)there was no significant difference between group D and E.Conclusion:(1)High cholesterol diet group performance:Myocardial arranging disorderly, partof fracture, mitochondria swell, cristae disorder,Compared with the respiratory chain,mitochondria complex activity decline, mitochondria ATP, coenzyme Q10content decrease,Tip high cholesterol can lead to cardiac muscle ultrastructure mitochondriachange and the mitochondrial energy metabolization obstacle;(2) atorvastatin regimen continued group performance:The myocardial fibers inorder, visible leap dish, slightly swollen mitochondria,With high cholesterol diet groupcomparison, respiratory chain, increased activity compounds, show thatatorvastatin regimen continued can reduce the myocardial structure damage, so as toimprove the respiratory chain of mitochondrial membrane complex activity;And betweenthe two groups, CoQ10mitochondrial ATP content difference was statistically significant,may and statins in the suppression of synthesis of cholesterol are inhibited the generationof CoQ10relevant.(3) atorvastatin regimen continued joint CoQ10group with atorvastatin regimencontinued group compared, mitochondria ATP, CoQ10content increased, show thatexogenous added can increase myocardial mitochondria CoQ10content, but alsoimprove the myocardial mitochondrial energy metabolization obstacle;Between the twogroups and respiratory chain, activity complex difference was statisticallysignificant, further verified mitochondrial structure damage on the influence of therespiratory chain complex activity.(4) common dose CoQ10group and high dose CoQ10group compared,mitochondria ATP, CoQ10content and respiratory chain compound active difference wasstatistically significant, may and dose group is too simple about, still need moreinformation with research. |
