Object:To discover proteomic define fingerprint which can be used to estimate the sensitivity of curative effect in treating colon cancer with FOLFIRI plan and explore the indications of using the drug by SELDIMethods:Collected Collected and detectfd the serum of66(40males,26females) colon cancer patients who had received surgery and FOLFIRI plan and had the observed curative effect before chemotherapy by SELDI-TOF-MS(surface-enhanced laser desorption/ionization time-of-flight mass spectrometry). The first stage, According to the Response Evaluation Criteria In Solid Tumor(RECIST,2000), the12patients were divided into2groups after application of FOLFIRI, stable group(6cases, SD) and inefficacy group(6cases, PD). Found the significant differences of the fingerprints between two groups with Biomarker Wizard Software. The second stage, selected54cases who had exact curative effect, verified the coincidence rate between the calibrated fingerprints and the actual curative effect according to the standard fingerprints obtained in the first stage.Results:(1) There were3significant different protein fingerprints between the two groups,M/Z were1372、1698、1865(p<0.05), baseline drift:±50H. Compared with the inefficacy group, M/Z:1372,1698were lower and M/Z:1865was height in the stable group.(2) In all patients, if the abundance of M/Z:1372±50H+<10, it may be attributed to the stable group, otherwise, it belonged to the inefficacy group, the coincidence rate between calibrated fingerprints and curative effect of the stable group was66.7%, which of the inefficacy group was71.4%. If the abundance of M/Z:1698±50H-<5, it may be attributed to the stable group, otherwise, it belonged to the inefficacy group, the coincidence rate with calibrated fingerprints and curative effect of the stable group was74.1%, which of the inefficacy group was81%; if the abundance of1865±50H+>10, it may be attributed to the stable group, otherwise, it belonged to the inefficacy group, the coincidence rate with calibrated fingerprints and curative effect of the stable group is70.4%, which of the inefficacy group is81%.Conclusion:It was feasible to estimate the sensitivity of curative effect in treating colon cancer with FOLFIRI Plan by SELDI. There were3significant different protein fingerprints between two groups,M/Z were1372.1698.1865. If the abundance of M/Z:1372±50H+<10,1698±50H+<5,1865±50H+>10,it may be attributed to the stable group, otherwise, it belonged to the inefficacy group.
