Objective:To detect the serum proteomic patterns in ovarian cancer patients,benign ovarian diseases and healthy women by using SELDI-TOF-MS protein chip array technology,screen biomarker candidates,build diagnostic models and evaluate its clinical significance.Methds:The Surface-enhanced laser desorption/ionization time-of-flight mass spectrometry technology(SELDI-TOF-MS)coupled with Weak-anion Exchange Chip(WCX2)was used to detect the serum proteomic patterns of 55 patients with ovarian cancer and 60 non-cancer diseases.We randomly selected 30 serum samples from patients with ovarian cancer and 25 serum samples from its control group,searched the differently expressed proteins by Biomark Wizard Software,set up diagnostic cast of ovarian cancer by Biomark Wizard Software and obtained its positive predictive value and accuracy and specificity.Result:190 protein peaks were detected at the M/Z values of 0 to 50000 in the training set(30 ovarian cancer and 30 control).A diagnostic pattern consisting of 4 protein peaks was established with a sensitivity of 90.0%(27/30)and a specificity of 86.7%(26/30).Among them,three protein peaks with 6879,8554,13726 were down-regulated,and one protein peaks with 11450 was up-regulated in ovarian cancer. The discriminatory pattern could recognize 21 from 25 cases of ovarian caner and 25 from 30 cases of control,achieving a sensitivity of 84.0%and specificity of 83.3%in the blind test for validation.Comparing the early stage with the late stage ovarian cancer sera,two protein peaks with 6148,6190 were detected in the early stage ovarian cancer,which were up-regulated.Conclusion:Using SELDI-TOF-MS technology,we have discovered a candidate ovarian cancer pattern consisting of 4 peaks that can distinguish serum samples from healthy controls and ovarian cancer patients.SELDI-TOF-MS is clinic significant for ovarian cancer early diagnosis.Combined use of bioinformatics tools and proteomic profiling provides an effective approach for screening potential serum tumor markers.
