Analysis On Differential Proteomic Fingerprints For Advanced Gastric Cancer Patients With LGT Fingerprints Changing

Object: To analyze the disposition characteristic of LGT fingerprints and the related fingerprints for advanced gastric carcinoma patients when the LGT fingerprints changing.Methods: SELDI and CM10 proteinchip was used to detect the serum protein fingerprints of advanced gastric cancer patients. After 1 year follow-up, all the patients were detected again by SELDI. According to the expression condition of LGT fingerprints (M/Z: 11000-12000), the patients were divided into 4 groups: A group: LGT changing from negative to positive; B group: LGT changing from positive to negative; C group: LGT fingerprints keeping negative; D group: LGT fingerprints keeping positive. And further more, 17 tumor patients were divided into 2 groups: 1 group and 2 group .To obtain fasting blood from 12 cancer patients of 1 group in the early morning, then to divide them into two samples after centrifugation: one sample is tested by SELDI immediately; another one will be tested by same way after one week's storage at - 80℃. To take the samples of blood from 5 cancer patients of 2 group at the two days before and after test respectively, at the same time, the samples should be tested by SELDI on the date the blood samples are obtained. This was the control group. The fingerprints of control group were influencing factor, they should be rejected. And the remaining fingerprints were LGT fingerprints and the related fingerprints. The different proteomic fingerprints were analyzed by Biomarker Wizard3.1 Software. And the predictive patterns were developed by Biomarker Pattern Software.Results: 1. Control group: 1 group: 1223 fingerprints were detected, among them, there were 39 significant differential protein peaks (p<0.05). M/Z: 1058,1085,1086, 1094,1008,1122,1011,1007,1030,1004,1005,1015,1017,1026,1028,1031,1033,1042, 1055,1056,1057,1066,1068,1069,1070,1072,1073,1123,1141,1144,1167,1029,1098, 1099,1100,1101,1102,1103 and 1109.2 group: 468 fingerprints were detected, among them, there were 18 significant differential protein peaks (p<0.05). M/Z: 753,1056,1058,1085,1008,1030,731,752,1007, 1017,1042,1057,1081, 1083,1099,1100 and 1124. In the significant differential protein peaks, M/Z: 1007, 1008, 1017, 1030, 1042, 1056, 1057, 1058, 1085 and 1099 expressed in two groups. It indicates the repetition and the stabilization of SELDI were not good.2. Analysis group: A group: there were 6 significant differential protein peaks (p<0.05). Compared with control group, there was no same fingerprint, and did not need to be rejected. The up- regulation fingerprints were M/Z: 11473, 11821, 11664, 11409, 11552 and 11947. There were no down- regulation fingerprints.B group: there were 14 significant differential protein peaks (p<0.05). Compared with control group, M/Z: 1098, 1029 and 1007 were same, and being rejected. Among the remaining fingerprints, M/Z: 878, 861, 874, 917 and 3264 were up- regulation, M/Z: 6523, 11509, 11669, 11413, 11351 and 6483 were down- regulation.C group: Only one significant differential protein peaks (p<0.05), it is M/Z: 1029. Compared with control group and rejected, there was 0 differential protein fingerprint. D group: there was 0 differential protein fingerprint (P < 0. 05).Conclusion: 1. In control group, M/Z: 1007,1008,1017,1030,1042,1056,1057,1058, 1085,1098,1099,731,752,753,1004,1005,1011,1015,1023,1026,1029,1031,1033,1041, 1042,1044,1055,1066,1067,1068,1069,1070,1072,1073,1081,1083,1086,1094,1101, 1102,1103,1109,1122,1124 were influencing factors and should be rejected.2. In analysis group, M/Z: 11473,11821,11664,11409,11552 and 11947 up-regulating were regard as the subtype of LGT when it changing from negative to positive; M/Z: 11509,11669,11413 and 11351 down-regulating were regard as the subtype of LGT when it changing from positive to negative; M/Z: 878,861,874,917 and 3264 up- regulating, and M/Z: 6523,6483 down- regulating were regard as the related fingerprints when LGT changing from positive to negative; and when LGT keeping negative or positive, there was no differential protein fingerprints.
