Background and PurposeGlioma is common malignant tumor, which are not accessible for surgery. In addition to high recurrence rates, it has the higher mortality. The current treatment strategies for malignant glima include surgical resection followed by radiotherapy alone or in combination with chemotherapy. Despite this multimodal therapy, the therapeutic effect is not achieved. Due to the carcinoma’s occurrence and development is a complicated multi-stage, multi-channel interaction process, studies begin to focus on the cancer gene therapy involved oncogene activation and anti-oncogene inactivation. So the further research of associated genes in the development of glioma and the exploration of the targeted gene become essencial.As a novel tumor suppressor gene, PDCD4plays an important role in the process of programmed cell death, in addition to imhibit the occurrence of transcription and translation. It is known that there a high deletion rate of PDCD4gene in the lung cancer, which is high related to the degree of malignancy. The low espression of PDCD4was also shown in the liver cancer. It becomes important to investigate of the espression of PDCD4and its relationship with the classification of tumor. PDCD5, a novel gene discovered by the Human Disease Genomics Center of Beijing University, with independent intellectual property rights, has been proved to be associated with apoptosis, with prompting the occurrence of apoptosis under the effect of inducing factor. It has been demonstrated that there is a low expression of PDCD5in leukemia and some other malignancy tumors such as clear cell carcinoma of kidney and ovarian cancer. What is the expression status of PDCD5in human glioma? What is its role in the glioma’s occurrencre and development? It is chosely associated with PDCD4in the process of glioma occurrence. To study the espression of PDCD4and PDCD5in the glioma sample, we use the RT-PCR to detect their mRNA expression level and the discuss the possible roles of PDCD4and PDCD5in the glioma’s occurrence and development. This may provide theoretical basis for searching for innovative therapies for glioma.Materials and Methods44glioma samples, including26males and18females(mean age38.19+18.32), were collected from the second clinical college of Zhengzhou University from January2010to October in2011, confirmed by pathology department after surgery. All the samples were kept with due care and all the patients with first attack, without chemoradiotherapy before operation. All the patients were divided into two groups according to2007WHO Classification of Tumours of the Central Nervous System, Grade I:7cases, Ⅱ:17cases, Ⅲ:11cases, IV:9cases.Low grade groups(24cases) including grade Ⅰ and grade Ⅱ, high grade groups(20cases) including grade Ⅲ and grade IV.12non-tumor tissues were considered as the control group. The mRNA expression of PDCD4and PDCD5genes were determined by RT-PCR in each sample. The results were analyzed with the statistics software of SPSS17.0and Probability values α=0.05were considered statistically significant.Results1. The mRNA expression of PDCD4The mRNA expression levels in the glioma、non-tumor tissues、low grade group、 high grade group were:0.547±0.204、0.939±0.134,0.631±0.233、0.468±0.184.There was significantly difference between glioma and the non-tumor tissues as well as between low grade group and high grade group.2. The mRNA espression of PDCD5The mRNA expression levels in the glioma、non-tumor tissues、low grade group、 high grade group were:0.870±0.186、0.688±0.176、0.746±0.202、0.555± 0.169.There was significantly difference between glioma and the non-tumor tissues as well as between low grade group and high grade group.3. The correlation between PDCD4and PDCD5The mRNA expression of PDCD4shows positive correlations with PDCD5.(r=0.583,P<0.05)Conclusion1. The mRNA expression levels of PDCD4and PDCD5in glioma tissue were both lower than those in the control group, with decreased with the upgrade of the pathological grade.2. The expression of PDCD4was positively correlated with PDCD5. Simultaneous enhanced expression of PDCD4and PDCD5prohibit the tumor growth and were closely associated with prognosis of glioma, Their expression were not isolated incidents.
