Objective: To detect the expression of PDCD4 mRNA and protein in human gliomas, and discuss their relationship between the genes and gliomas.Methods: Real-time quantitative reverse transcription PCR(RT-PCR) and immunohistochemistry(FIS) was performed on glioma surgical specimens and normal brain specimens. Statistical analysis were used to interpret the relationship between the expression and gliomas.Results: The expression of PDCD4 mRNA in gliomas was lower than that in normal brain tissues(P<0.05),and the expression of PDCD4 mRNA in gliomas of gradeâ… -â…¡was much higher than that of gradeâ…¢-â…£(P<0.05). PDCD4 protein expression level in glioma tissues was lower than that in normal brain tissues by means of immunohistochemistry, and gradeâ… -â…¡expressed higher levels of PDCD4 protein than gradeâ…¢-â…£.Our data suggest that the loss of PDCD4 expression correlate with the pathological grade and clinical features of the glioma significantly. The follow-up results showed the prognosis of the patients whose PDCD4 stained positive by immunohistochemistry was better than those stained negative. COX multivariate regression analysis showed that the protein expression lever of PDCD4 and the pathological grade were the most important significantly independent prognostic factors for glioma patients.Conclusions: PDCD4 mRNA and protein levels are both lowly expressed in human gliomas, which may play an important role in the oncogenesis and progression of human gliomas. And the expression of PDCD4 mRNA and protein in gliomas of gradeâ…¢-â…£is much lower than that of gradeâ… -â…¡,which demonstrate that PDCD4 is relevant to the malignant level of glioma. The new method targeting the PDCD4 can be the criterion of the malignant level of gliomas. It may exert a crucial role in the prognosis judgement of gliomas in clinic.
