PDCD5 accelerant apolosis, whose gene is 6kb, locus on 19q12-q13.1, which composed by 6 exon and 5 intron. PDCD5 express on more than 50 kinds of tissue of human. It highly express on adult's heart,testis,kidney,adrenal gland and placenta, low express on embryonic tissue. PDCD5 accelerant apolosis by regulation caspase-3. PDCD5 nuclear translocation maybe is a signal of apoptosis. Survivin suppressor apotosis, whose gene is 14.7kb, locus on 17q25, which composed by 4 exon and 3 intron, comprised 2 transcription initiation point, the upstream of transcription initiation point is 2560-2920, where defect of TATA promoter and GCGC, coding 142 kinds of amino acids. Under physiology circumstances, Survivin protine unexpress on human's majority tissue except placenta and thymus, but majority malignant tumor express Surviving.Investigation have indicate that Survivin can inhabit cytosis, promote cell proliferation, accommodation cell caryocinesia embryonic development and vasoformation.This investigation detect PDCD5,Survivin and mRNA express level which is utilized by immunohistochemistry and PT-PCR on normal colon tissue and colon carcinoma tissue, discover the relation of PDCD5,Survivin and colon carcinoma, to find a new management target of colon carcinoma's early diagnosis and management.Provide a theory evidence of gene therapy on colon carcinoma. Expression of PDCD5 and survivin protein in 58 colon carcinoma tissues and 14 normal colon tissues were detected by Ultrasensitive Streptavidin-Peroxidase immunohistochemistry staining. Expression of PDCD5 and survivin mRNA in 58 colon carcinoma tissues and 14 normal colon tissues were detected by Reverse Transcription Polymerase Chain Reaction (RT-PCR). Associate with clinical pathological parameters to analysis the relationship of PDCD5 and Survivin with colon carcinoma.Results:1,PDCD5 protein was detected 19 out of 58 cases colon carcinoma tissue (32.7%) and 12 out of 14 cases normal colon tissue (85.7%). PDCD5 mRNA was detected 20 out of 58 cases colon carcinoma tissue (34.5%) and none of the normal colon tissue.The expression of PDCD5 mRNA had significant difference between colon carcinoma tissue and normal colon tissue (P<0.05).2,The positive rate of Survivin protein in colon carcinoma tissue was 56.8% (33/58) and 28.5% (4/14) Was detected in normal colon carcinoma. The positive rate of Survivin mRNA in colon carcinoma tissue was 58.6% (34/58) and 28.5% (4/14) was detected in normal colon tissue.The expression of Survivin mRNA had significant difference between colon carcinoma tissue and normal colon tissue (P<0.05).3,The expressions of PDCD5 protein and mRNA were correlated with differentiation and lymph node metastasis. There was no relation with age,gender,the depth of invasion and clinical stages. As the decrease of differentiation, the expression rates of PDCD5 protein and mRNA were decreased.The difference between highly and lowly differentiated colon adenoma had a statistical significance, the neutral d ifferentiated and lowly differentiated colon adenoma had a statistical significance, but the highly and the neutral differentiated colon adenoma had no statistical significance (P<0.05). Comparing to without lymph node metastasis, the positive rate of Survivin protein and mRNA in colon carcinoma tissue with lymph node metastasis was lower. 4,The expressions of Survivin protein and mRNA were correlated with differentiation and lymph node metastasis. There was no relation with age,gender,the depth of invasion and clinical stages. As the decrease of differentiation, the expression rates of Survivin protein and mRNA were increased. The difference between highly and lowly differentiated colon adenoma had a statistical significance,the neutral differentiated and lowly differentiated colon adenoma had a statistical significance, but the highly and the neutral differentiated colon adenoma had no statistical5 significance (P<0.05). Comparing to without lymph node metastasis, the positive rate of Survivin protein and mRNA in colon carcinoma tissue with lymph node metastasis was higher.5,PDCD5 and Survivin had a negative correlation in colon carcinoma.Conclusion:1. Comparing with normal colon tissue, the positive rates of PDCD5 protein and mRNA were downregulated in colon carcinoma. It demonstrate that promote apoptosis resulted from abnormal expression of PDCD5 had an important significance in the occurance of colon carcinoma. Decrease of expression rate has a great prognostication of early diagnosis in colon carcinoma.2. The express rate of Survivin in normal colon tissue was low level, but the expression in colon carcinoma tissue is strong. So abnormal expression of Survivin may be have effect on pathogenesis of colon carcinoma.3. The expressions of PDCD5 and Survivin were correlated with differentiation and accompanied lymph node metastasis. As the dereasing of differentiation and having lymph node metastasis, the expression of PDCD5 was downregulated, but the the expression of Survivin was upregulated. Combine detecting the expressions of PDCD5 and Survivin may be used to estimate the degree of malignance and prognosis of colon carcinoma.
