Abstract/Summary:
Objective: To investigate the effects of intraperitoneal injection of CpGoligodeoxynucleotide (ODN) on the expression of Mucin5AC (MUC5AC) in lungtissue and the levels of IL-33in BALF in asthmatic mice, preliminary evaluate thetherapeutic effect of CpG ODN on mucus hypersecretion in airway, and affirm thechanges of the level of IL-33in asthmatic mice.Methods: Twenty-one BALB/c mice were randomly divided into three groups,including normal control group, asthma model group and CpG ODN treatment group,seven mice per group. Mice in asthma model group and CpG ODN treatment groupwere sensitized and challenged with ovalbumin (OVA), and normal control groupwith saline instead, mice in CpG ODN treatment group were treated by intraperitonealinjection of CpG ODN one hour before challenged. Total cells and eosinophilpercentage (EOS%) were checked in bronchoalveolar lavage fluid (BALF); thepathomorphological changes in the lung were observed by hematoxylin-eosin staining(HE), the goblet cells of bronchi were observed by AB-PAS staining; the expressionof Mucin5AC (MUC5AC) was detected by immunohistochemical staining; the levelsof IL-33in BALF were determined by ELISA.Results: Compared with asthma model group, the total cell number and EOS%inBALF were significantly decreased, the hyperplasia of goblet cells was remarkablyrelieved, the expression of MUC5AC in lung tissue was decreased in CpG ODNtreatment group. Compared with normal control group, the concentration of IL-33inBALF of asthma model group was significantly increased (P<0.05), and theconcentration of IL-33in BALF of CpG ODN treatment group is higher than that ofasthma model group (P<0.01). Conclusion: Intraperitoneal injection of CpG ODN can effectually relieve the allergicairway inflammation and inhibit the hyperplasia of goblet cells in asthmatic mice,CpG ODN may play a role in the treatment of mucus hypersecretion in airwaythrough down-regulating of MUC5AC. The levels of IL-33in BALF were increasedin asthmatic mice, IL-33may play a crucial role in the pathogenesis of asthma, butCpG ODN can induce the expression of IL-33, IL-33may interfere with thetherapeutic action for CpG ODN on asthma.
