Effects Of Complex Prescriptions For Replenishing Qi And Resolving Phlegm On Goblet Cell And Muc5ac Of Asthma Mice Airway

Purpose:To explore the mechanism of Yiqi Huatan Decoction for inhibiting asthmatic airway inflammation, and reducing the secretion of airway mucus by observing the changes of the level of the lung mucin (MUC5AC), ECP and 1L-8, and the effects of Yiqi Huatan Decoction.Method:40 SPF BALB/c mice were randomly divided into normal control group, asthmatic group, the western medicine group (prednisone acetate) and the Chiniese medicine group(Yiqi Huatan Decoction). Mice were established.into asthmatic model via sensitizing and challenging by ovalbumin. The traditional Chinese medicine group and the western medicine group which were challenged 30min ago were started to be treated by Yiqi Huatan Decoction and prednisone acetate for 10d, respectively.Pulmonary histological changes were assessed by the lung tissue hematoxylin-eosin (HE) staining and airway goblet cell changes by periodic acid Schiff (PAS) staining, Additionally,the level of the lung mucin (MUC5AC) was determined by ELISA.Resumice:1. The effects of Yiqi Huatan Decoction for infiltration of inflammatory cells in lung tissue of asthmatic mice.It was indicated that compared with normal control group, a large number of inflammatory cell infiltration were observed in asthmatic group by HE staining in the lung tissue. Especially around small airway and vessles, mucosal edema and epithelial hyperplasia were obvious, Which, in comparison, were significantly reduced in both treatment groups.2. The effects of Yiqi Huatan Decoction for goblet cells in lung tissue of asthmatic mice. It was showed that PAS positive staining cells were uncommen appeared in airway mucosa epithelium in normal control group by PAS staining. However, lots of PAS positive staining cells were seen in asthmatic group. The level of PAS positive staining cells in both treatment groups was significantly lower than asthmatic group, in addition, asthmatic group by TCM treated was better than asthmatic group by western medicine.3. The effects of Yiqi Huatan Decoction for MUC5AC in lung tissue of asthmatic mice.It was displayed that the level of MUC5AC in lung tissue was 26.46±5.10 ng/ml in normal control group, and was 54.79±5.43 ng/ml in asthmatic group by ELISA. In the asthmatic group, the level of MUC5AC in lung tissue was significantly higher than the control group(p< 0.05),and was significantly lower than treatment groups (the level of MUC5AC in lung tissue was 36.14±5.82 ng/ml in asthmatic group by western medicine, and the level of MUC5AC in lung tissue was 26.14±6.91 ng/ml in asthmatic group by TCM treated). Besides, asthmatic group by TCM treated was better than asthmatic group by western medicine(p<0.05).4. The effects of Yiqi Huatan Decoction for IL-8 in lung tissue of asthmatic mice.It was suggest that IL-8 in lung tissue was 256.09±32.05 pg/ml in normal control group,647.18+37.78 pg/ml in asthmatic group,455.35+72.36 pg/ml in asthmatic group by western medicine, and 445.03±50.16pg/ml in asthmatic group by TCM treated. Compared with those in the normal control group, the expressions of IL-8 in lung tissue was significantly increased in asthmatic group (p< 0.05),and significantly decreased in treatment groups(p<0.05). 5. The effects of Yiqi Huatan Decoction for ECP in lung tissue of asthmatic mice.The results showed that the level of ECP in lung tissue was 19.22±3.20 ng/ml in normal control group, and was 31.55±6.27 ng/ml in asthmatic group. Compared with those in the normal control group,the expressions of ECP in lung tissue was significantly increased in asthmatic group (p<0.05). The expressions of ECP in lung tissue asthmatic group by TCM treated was 22.32+2.58ng/ml, and in asthmatic group by western medicine was19.50±5.40ng/ml. It was significantly decreased in asthmatic group by TCM treated(p<0.05). In addition, there was no marked difference between asthmatic group by western medicine and asthmatic group by TCM treated.Conclusion:1. In the asthmatic group, the infiltration of inflammatory cells, and airway goblet cell hyperplasia were displayed.2. In the asthmatic group, the the'level of MUC5AC protein, ECP,IL-8 in lung were increased.3. Yiqi Huatan Decoction can improve airway inflammation and reduce goblet cell hyperplasia and MUC5AC,ECP,IL-8 expressions.
