The probiotic is a preparation to contain a certain quantity, composition and specific microbial agents or products, these micro-organisms through the promotion of the host micro-ecological balance, and beneficial to host health.Can improve the host intestinal flora ecological balance in the use or enhance its non-specific immune function indirectly to promote animal growth and feed conversion ratio, to improve the health and health of the host role.The study uses a combination of traditional bacteriological identification methods and molecular biological methods, were identified in our laboratory isolated from the carp body and save a probiotic Bacillus A7.And then, it studied the best solid state fermentation, the feeding protein degradation in vitro and the feeding effects on Jian carp of the strain.The probiotic strain A7was identified by using16SrDNA sequence analysis method. The conserved general primers of bacteria16SrDNA were designed according to the literature. The genomic DNA of A7was extracted and amplified the target16SrDNA sequence. Meanwhile, the sequence was compared with the BLAST database of NCBI. Then, it used the biology software clustalx1.81and mega4.0to construct the phylogentic tree. At last, combined the bacteriological identification; A7strain was identified as Bacillus subtilis. On the basis of single factor optimization result, the best solid fermentation conditions were confirmed. The conditions were:water ratio1:1,inoculum concentrations%,temperature28℃and capacity ratio16%. Solid medium in accordance with the best solid state fermentation conditions for fermentation production of power, powder number of viable cells of1.0X109cfu/mLThe feeding protein degradation in vitro was studied by using orthogonal design text. The text showed that the best temperature which A7strain degraded the bean pulp, rapeseed dregs and corn flour were28℃,30℃and25℃; the best inoculums was5%;the best water ratios were1:1.2,1:0.8and1:1.2, the best fermentation time were72h,72and48h. In the orthogonal design text, after fermentation degradation, the DH of the bean pulp, rapeseed dregs and corn flour text group were20.92%,7.12%and8.81%, respectively, compared with the control group to improve12.68%,2.95%and3.81%. And the ALPC were11.86,38.11and11.47, lower than the control group about48.83%,25.09%and3.61%.From the result of feeding Jian carp, added to the feed to0.5%,1.0%and1.5%Bacillus A7strain experimental group Jian carp weight gain than the control group were increased by25.60%,69.07%and36.03%, add commercially available Bacillus subtilis powder test the formation of carp weight gain rate than the control group increased by15.07%; hepatopancreas amylase, intestinal protease and amylase, respectively, than the control group increased by2.05%to11.49%,10.52%to31.44%and3.94to9.82%(P<0.05). Among them, feed conversion ratio of the minimum dosage of1.0%A7strain of the experimental group,1.55,32.90%lower than the control group(P<0.05), the efficiency of dietary protein increased by48.55%compared with control group(P<0.05).
