In this study, the corn cob was used. Using Pleurotus eryngii as testing material, effect of the corn cob content and different auxiliary materials on the growth conditions, agronomic trait, yield and biological efficiency of Pleurotus eryngii were investigated, obtaining the optimal combination of the corn cob and auxiliary materials. Under the optimum culture formula, five strains were vaccinated to Pleurotus eryngii to obtain the optimal strain, judging by several indices like yield, biological efficiency, growth cycle and quality etc.. The aim is to provide the theoretical evidence for standardized and massive culture of Pleurotus eryngii. The results were shown as follows:1. Screen of auxiliary materials. Different auxiliary materials combining with60%corn cob were applied to culture Pleurotus eryngii. According to the growth condition, the agronomic trait, yield and biological efficiency of Pleurotus eryngii, the optimum auxiliary materials were: cottonseed hull and bran. Under the optimal conditions, the growth rate of Pleurotus eryngii reached as high as4.8mm/d, and the yield and the biological efficiency were also the highest, being75.6g/bottle and60.5%, respectively.2. Screen of corn cob. Based on above, with cottonseed hull and bran as the auxiliary material, the corn cob of50%,60%,70%and80%were used to gain the optimal content of corn cob. As the results showed, the70%corn cob was the optimum culture materials, under which, the growth rate, yield and the biological efficiency of Pleurotus eryngii were4.8mm/d,77.1g/bottle and61.7%, individually. In terms of the variance analysis, the difference was significant, compared with other treatments. In conclusion, the optimal culture conditions were:70%of corn cob,14%of cottonseed hull,9%of bran,3%of corn powder,1%of sμgar,1%of lime powder,1%of gypsum and1%of monopotassium phosphate.3. Screen of strains. Under the optimum culture conditions mentioned above, the strains named nongxing, cixing, taixing, xuerong and xing-5were vaccinated to Pleurotus eryngii respectively. The growth rates of five strains were not significant different. The yields of nongxing and cixing strains were higher, and the biological efficiencies of the two were58.5%and56.8%respectively, which was significant different from other treatments. The volume-weight and polysaccharide of nongxing and xing-5strains were significant higher than those of others. Generally, the nongxing strain with higher growth rate, volume-weight, yield and polysaccharide, were the optimal strain.4. Comparison of liquid strain and solid strain. The screened corn cob and nongxing strain were inoculated to the liquid and solid strain respectively, investigating the characters and indices of the hyphae and sporocarp of Pleurotus eryngii. As known from the results, under the different strains, both of the hyphae were compact and pure white, and the volume-weight, yield and polysaccharide content of the sporocarp did not vary a lot. In comparison with the growth rate of hyphae, liquid strain was better. Cultured in the liquid strain, the hyphae grown quickly, which could shorten the culture cycle and save the cost for the scale production of Pleurotus eryngii.
