With the development of molecular biology, molecular marker technology has been widely used in the identification of the purity of the various crops. Our members have selected3SSR makers which can distinguish some varieties of Brassica napus hybrids from their parents, but did not do the appropriate field identification comparative analysis. This experiment aims to compare the field identification results with SSR markers identification results to determine identification accuracy of the above-mentioned3markers.We tested the purity of12copies of hybrid canola seeds with3SSR makers developed by our lab, than record the rate of paternal, rate of sterile plants, rate of fertile plants and purity. At the same time, select12copies hybrids whose numbers are form1to12, than planted in the field. We record the rate of fertile plants in the florescence, and compared the SSR marker identification results with field identification results. Making the paired data t-test and regression analysis of the two results by two kinds of identification methods of12copies samples whose numbers are form1to12. We make the analysis of variance of9copies samples whose numbers is form4to12, trying to find the difference between the two methods.Main results:1. Purity identification of12copies of rape seeds using SSR maker. The result shows that most samples are qualified. The overall pass rate is83%. Except for sample number2, other samples containing the paternal hybrid strains, and paternal ratio is less than the sterile strain ratio, it indicating that it is the sterile plants rate mainly lead to low purity of hybrid.2. Compared the SSR marker identification results with field identification results of12copies samples whose numbers are form1to12. The average rate of fertile plants was92.00%by SSR identification, and94.13%by field identification. The field average rate of fertile plants was higher than the SSR identification results. Making average rate paired data t-test. It showed that there is no difference between the two methods. 3. After regression analysis, we get a linear regression equation between the two data: y=56.4x+42.2(t=3.28**), we can estimate the value of the field identification by the linear regression equation.4. Variance analysis of field identification results showed that, there is significant difference between the pair9A×2-3and9A×2-7, and very significant between the pair9A×2-3and11A×2-1. There is difference between the3pairs. Variance analysis of identification results using SSR makers showed that, there is significant difference between the pair9A×2-3and11A×2-1.
