Seed Purity Identification And Finger-print Construction Of New Wheat Variety Yunong186

Purity identification and molecular fingerprints construction of new wheat varieties is of great significance for screening germplasm,counterfeit purification and protecting the rights and interests of new varieties.Yunong 186 is a new wheat variety bred by Henan Agricultural University and released by Henan Province in 2017.In this study,the purity of the new wheat variety Yunong 186 and the differences between its sister lines were identified by field morphology and molecular markers,and 18 SSR markers with good polymorphism were screened for constructing their fingerprints,which provides basis for its seed detection and protection of the variety rights.The results are as follows:1.The difference of agronomic traits between 30 seed samples and standard samples of Yunong 186 was mainly reflected in plant height,neck length and grain number,and the number of spikes per plant,fertile spikelets number,sterile spikelets number and panicle length were smaller.Significant difference analysis showed that thee samples might have confounding,and further molecular markers analysis showed that one of the samples was mechanically mixed with one single plant.2.The difference of agronomic traits between the 13 sister lines and the standard sample of Yunong 186 was mainly found in plant height and neck length of the main stem,and small differences in other traits.Molecular marker Xwmc622 can distinguish the difference between four sister lines and the standard sample,and the number of marker is too small to distinguish all the sister lines and the standard sample3.Using molecular marker for purity detection,we can mix five seeds or equal amount of leaves to extract DNA,which can reduce 4/5 of the workload.4.The genetic differences of 31 wheat cultivars were analyzed by 18 SSR markers,and the molecular fingerprint of Yunong 186 were preliminarily constructed.A total of 101 polymorphic loci were detected,with 4 to 9 loci per pair primers,with an average of 5.60.The PIC(Polymorphism index)values of these markers varied from 0.42 to 0.74 with an average of 0.65.The combination of two pairs of primers Xgwm333 and Xgwm285 can distinguish Yunong 186 from the other 30 wheat varieties.
