The Studies On Purity Dentification Of Rapeseed Hybrids Shaanyou No.10

Seed purity is the important index of plant,and infact the quantity and quality directly and obviously.Various techniques for seed purity identification such as morphological marker,microscopic observation,seed storage protein,isozymes,RFLP,RAPD,AFLP and SSR in seed testing are reviewed on this report;and assessed all the methods about purity testing objectively and justified.The identification of rape hubrids Shanyou No.10 was made in plant in the field,seed storage protein and isozyme.The results are obtained as follows:1. Microscopic observation on anther development process can make differences from normal plant and sterile plant.The result shows that,the growth anther of sterile plant is inhibited at the stage of chear sporial cell,and it could not form normal archesporial cell and pollen sac.Male sterile line"208A"is inhibited at the stage of chear sporial cell.The characters is that the anther is formed of wall tissue and vascular bundle.It does not differentiate the sporogenous cell and the pollen sac.So it belongs to non-sporarge lines.The method doesn't fit to purity identification.2. Hyprids Shanyou No.10 are investigated in the field,and make of normal plants,sterile plants and mixed plants,and calculate the rate of sterile plant and mixed plant.The result is,the number of the plants is 1150,the number of normal ones is 1051,the number of sterile ones is 96,and the number of mixed ones is 3.The final result is that the purity is 91.39%.3. The purity of Shanyou No.10 is analyed by EST-isozyme and POD-isozyme.The result showed that,EST-isozyme can identify the male line from the F1 generation,but not identify the female line from the F1 generation;POD-isozyme can identify them effectively,hybrid F1 and its parents are produced its own specific bands.The purity is 88.94%,as the same as the resule in the field.
