In this study, eight varieties of sheep and goats with congenital immune parameters weredetermined, in which four goat breeds such as Laiwu black goat, Laoshan dairy goat, Boergoat, Lubo goat, total of176,and four sheep breeds such as Small Tail Han sheep, Aohanfine-wool sheep, Poll Dorset, Suffolk, total of200. Jugular vein blood were divided into twolots, one of which, anticoagulanted by EDTANa2,were used to detect the routine blood(WBC, LYM LYM%, MID, MID%, GRAN, GRAN%, RBC, HGB GCT, MCV, MCH,MCHC, RDW-SD) by Automated hematology analyzer. The sera, collected from the othervein blood, were used to detect the IL-1, IL-2, IL-6, IL-18and IFN-γ, NK cells, LYS, byELISA-kit. Treat species, breed, age, species source as the dependent variable, with themethods and principles of biometrics, to contrast between physiological biochemical constantand immunological constant.First, we used species as dependent variable to do independent T-test. Results were asfollows: To the immunological parameters, the IL-6and LYS level of goats was significantlyhigher than sheep (P<0.01), while the IL-1, IL-18, and NK cells level of sheep wassignificantly higher than goats (P<0.01). But IL-2and IFN-Y levels are not significantlydifferent between the two species (P>0.05). To physiological indicators, the MID, GRAN,GRAN%, HGB, MCH and HCHC level of goats was significantly higher than sheep (P<0.01),while the WBC, LYM, LYM%, MID%, RBC GCT, MCV and RDW-SD level of sheep weresignificantly higher than goats (P<0.01).Second, breed source (local breed and introduced breed) was used as dependent variable todo independent T-test. Results were as follows: to goats, the NK and LYS level of local breedis higher than introduced breed (P<0.05); and the WBC, MID%, MID, RBC, HGB, GCT,MCV and RDW-SD level of local breed was higher than introduced breed (P<0.05);especially, the WBC, RBC, HGB, and MCV level of local breed was significantly higher thanintroduced breed and has significant difference (P<0.01). To sheep, the IL-1, IL-2, IL-6, IL-18,IFN-gamma, NK and LYS level of introduced breed was significantly higher than local breed(P<0.01); but, the WBC, LYM, MID, GRAN, RBC, GCT and MCV level of local breed wassignificantly higher than introduced breed (P<0.01).Third, four varieties of goats and sheep were as dependent variables to do a single factor ANOVA multiple comparison analysis. The results were as follows: to four goat varieties,Laiwu black goats have a higher level of NK and LYS than Laoshan dairy goat, Boer goat andLubo goat, but the difference was not significant. Although Boer goat has a significantlylower level of IL-1, IL-2than the other three, but the difference was not significant, too. Thelevel of IL-18and IFN-gamma has no significant differences among the four varieties. Laiwublack goats has a significantly higher level of WBC, LYM, GRAN, RBC GCT, and MCV thanthe other three species (P<0.01). To four sheep varieties, Suffolk sheep has the highest level ofIL-1, IL-6, IL-18, IFN-Y, LYS and the difference was very significant (P <0.01). Small TailHan sheep has the lowest level of IL-18, IFN-γ and the difference was very significant(P<0.01). The Aohan fine-wool sheep has a higher level of WBC, LYM, MID, RBC GCT,MCV, and RDW-SD water than those in the Poll Dorset and Suffolk sheep, and the differencewas significanted, but the difference between the Small Tailed Han and Ao Han fine-woolsheep was not significanted, and the same to the difference between Poll Dorset sheep andSuffolk sheep (P>0.05).Forth, the age of goats and sheep was as dependent variables and used to do independentT-test. Results were as follows: as to Laiwu black goat, the IL-6, IL-18, and NK level of adultsheep was higher than those of young sheep, while other indicators have no significantdifferences (P>0.05); Laoshan dairy goat, the IL-1and IL-2level of adult sheep was higherthan young sheep (P<0.01), and no significant differences among other indicators; Boer goat,the LYS level of young sheep was higher than the adult sheep (P<0.01), and no significantdifferences among other indicators (P>0.05); regarding to Small Tail Han sheep, the IL-1,IL-2, NK and LYS level of adult sheep was higher than the young sheep (P <0.05), and nosignificant differences among other indicators (P>0.05). To the age factor, there were nosignificant differences among Aohan fine-wool sheep, Poll Dorset and Suffolk sheep (P>0.05).Physiological indicators: to Laiwu black goat, the young sheep was with a higher level ofWBC, LYM and MID than the adult sheep; to Boer goat, the young sheep was with a higherlevel of MID than the adult sheep; to Dairy goat there was no significant difference with theage (P>0.05). Small Tail Han sheep, the WBC, LYM, MID, RBC GCT, MCV, and RDW-SDlevel of adult sheep was significantly higher than young sheep (P<0.01). The Aohan fine-woolsheep, RBC, MCV level of adult sheep was higher than young sheep. Poll Dorset and Suffolksheep, there was no significant difference with age (P>0.05).These results suggest that: Local goat varieties of the innate immune levels weresignificantly higher than the introduced species. Introduced species of sheep in theimmunological parameters were higher than local varieties, indicating its varieties advantages, lower than the local varieties in the blood physiological parameters. Between the variousvarieties of the goats, Laiwu Black Goat is a highest leve. Immunological indexes level ofSuffolk sheep was the highest. In blood physiological indicators, Small Tail Han sheep andAohan were higher than Poll Dorset and Suffolk sheep. In innate immune indicators, the levelof adult sheep was higher than the young sheep. In blood physiological indicators, the level ofyoung sheep was higher than the adult sheep.Through the above research, we provide reasonable experimental data to build theevaluation system of sheep innate immune level, which was under the natural state. Thisprovides a theoretical basis for future breeding of sheep varieties, especially for the newvarieties of disease-resistant sheep as well as drug development.
