Agrocybe aegerita, is part of Eumycota, Basidiomycetes, Agaricales, Bolbitiaceae. Agrocybe aegerita is a kind of common edible mushrooms, but the acclimatization effect still can not be like the type of Lentinus edodes, and still need to for more research. Now edible fungus agronomy traits research is uneven, Agrocybe aegerita’s agronomy traits occasionally reported, although not comprehensive, but still as a good reference. Therefore, this experiment is the contrast of biological efficiency and growth traits which is directly related to the properties of production of properties and characteristics. Moreover, based on the chaos of Agrocybe aegerita’s classification, edible fungus market lacks of unified management, strains management confusion always happen occasionally, in order to strengthen the management of the Agrocybe aegerita strains, it is particularly important that Agrocybe aegerita strains identify rapidly. So during its research, the germplasm classification of genetic marker is becoming more and more important.In this paper,17Agrocybe aegerita strains’s many agronomy traits which are mycelium growing, color of sporophore, stress resistance, difficulty of pileus-open, yield, biological efficiency and so on, are used in comparison experiment. Most of strains have very good mycelium growing, easy pileus-open, strong stress resistance. Their primordium appearance used about34-48days, first fruiting spent40-53days, biological efficiency nearly50%above. Terra brown Strain has better growth vigor, but White-mushroom and As-F’s agronomy traits are weakly. After comprehensive comparison, we selected As-1which is the fine core of the relative strain.For the development of EST-SSR markers in Agrocybe aegerita and its application in characterization of different mushrooms,40pairs of primers were designed based on the expressed sequence tags obtained from the shiitake Lentinula edodes. The results showed that13of28selected primer pairs could reveal polymorphisms among Agrocybe aegerita varieties, which accounted for46.4%of primers selected. And further verified the transferability of EST-SSR primer. Amplified PCR product has a total of81polymorphic bands, the number of bands’ range is2-14, with an average of5.86bands, amplified fragment length is between100-400bp. The genetic similarity coefficient of17Agrocybe aegerita strains a range of0.633to1.000. Polymorphism results of the PCR products can be successfully classify, reflecting the genetic differences between strains.Analyzed and compared the analysis of agronomic traits and the results of EST-SSR molecular markers. Among them, white mushrooms and As-F are two white Agrocybe aegerita, which genetic relationship is different from the other colors of Agrocybe aegerita, in line with the morphological differences. There are high similarity in the production and genetic similarity coefficient among from As-1, Guangchang F1and Guangchang F5.This will help more comprehensive analysis and theoretical studies the various features of Agrocybe strains. |
