There are more pharmacol-effects for fungal polysaccharide, such as immunomodulation, lower blood sugar, anti-stress, anti-tumor, antivirus, antiradiation, antioxidation etc, so they are used of medicines and functional foods. the research of fungi polysaccharides has caused great interests of scholars from home and abroad, their important biological effects have become the hotpoint of today's research.Agrocybe aegerita has dense aroma and delicious taste ,bundant nutrition and many for medicinal ;At present about reseacher of mushroom,but letter of Agrocybe aegerita . In this paper, in order to compare extraction method and their active functions, in order to provide the experimental bases to protect the cherish mushroom and to make it in dustralized.The extraction methods of Agrocyb Aegerita Polysaccharid including hot water extraction, Microwave-assised,Ultrasonic-assised, and cellulase method were investigated.The optimal extracting conditions for hot water extraction technology of extracting Agrocybe aegerita Polysaccharid by hot water: The research for optimization of extraction temperature, ratio of solid-liquid, the duration of time and the extraction of times in extraction polysaccharides from exopleura of ginkgo biloba, has been under taken by orthogonal experiment of L16(43). The extracting rate was 42.51%. The optimal technological conditions are as follows: temperature 90℃, ratio of solid-liquid 1:20, the duration of time 4h and the extraction of times 3 time. The optimum extracting condition microwave is power 320W, 6.0min. The extracting rate was 58.65% . The optimum extracting condition in ultrasonic field is frequency 59kHz, 20 min. The extracting rate was.68.83%.So those the dosage of cellulose enzyme, enzymolysis time, enzymolysis temperature and concentration of raw materials, which are four mainly factors of impacting the cellulose enzyme action effect to determine the optimum conditions of Agrocybe Aegirit Polysaccharid extraction by the methods of enzymatic + hot water extracting. The test results showed that: Optimum conditions of Cellulose enzyme + hot water extraction is: enzyme dosage 0.08%, enzymatic time 4h, enzymatic temperature 45℃, concentration of raw materials (volume ratio) 1:20, hot water extracting 4 hours, extracting 2 times. The extraction rate of tea mushroom polysaccharide is 57.62%.The scavenging effect of Agrocyb Aegerita polysaccharide on hydroxyl radicals(·OH)and oxygen radicals(O2·)were tested by salicylic acid method. Studying inhibition effect on Silkworm oils oxidation system by thiobaribituric acid(TBA), The result showed that Agrocyb Aegerita Polysaccharid can detcting hydroxyl radicals(·OH)and oxygen radicals(O2-·) deeply but poorly to lipid peroxide; their antioxidative activities should ofer scientific basis for their application. |
