In this paper we studied micropropagation technology of Guzmania ‘Denise’systematically, and established a better regeneration system combining with shoot tipdetoxification and high temperature detoxification. The results showed:1. Shoot tip detoxification culture: There were higher contamination rate and slowergrowth rate if we selected the shoot tip with the naked eye. In ultra-clean work bench, with adissecting microscope we took0.1-0.3mm shoot tips, the adventitious buds induced was betterand the survival rate could be more than90%.2. Higher temperature detoxification culture: In order to detoxification culture, weputting the tissue culture seedlings into the artificial incubator with high and low temperaturechanges. The higher temperature was38℃for12h/d, and the lower temperature was28℃.Theexposure time was12h/d and the light intensity was5000-6000lx. After30days treatment, theseedlings would be taken the growing point again under the microscope. Regenerated plantsafter higher temperature treatments growed significantly more quickly than untreated tissueculture.3. The best adventitious bud induction medium was ZS+6-BA1.0+IAA0.5+TDZ0.02+AC0.5. In this medium, the explants browning rate grew faster. Callus induction medium wasZS+6-BA1.0+IAA0.5+TDZ0.02+NAA0.01+CPPU0.1. In this medium, the shortening stemsbegan to expand after45days. The callus differentiation medium was ZS+AC0.5. The bestrooting medium was ZS+IAA1.0+AC0.5. This study shows that in order to prevent Guzmania‘Denise’ growing pause, we should avoid the use of NAA and IBA which was easy to producecallus.4. The best propagation model was: optimal plants selection-shoot tip detoxification-higher temperature detoxification-shoot tip detoxification-callus induction and proliferation-callus differentiation-rooting-transplanting.
