European and American poplar 107?Populus x euramericanacv."Neva"?is the Yang Sh uxin variety introduced from Italy in 1984 by researcher of Chinese Academy of forestry,wh ich has the advantages of tree shape,dry shape,small cutting degree,small crown,high quali ty material,less disease and insect harm,rapid growth and so on.It has become the north of China.One of the fine varieties of widely cultivated poplar.In recent years,the emergence o f many fungi,bacteria and viruses caused the emergence of many fungi,bacteria and viruses due to the long-term use of asexual reproduction in European and American poplar 107,whi ch made the 107 species of poplar in Europe and America slowly degenerate,plant resistanc e to stress and rapidly decrease the speed.In order to restore 107 fine varieties of Euramerica n poplar and rejuvenate their seedlings.In this study,we used the stem segments,stem tips,l eaves,roots and the stem segments,buds and vase leaves of the 107 fields of poplar in Europ ean and American poplar as experimental materials.In addition,2 strains of 107 Euramerica n poplar were selected for tissue culture and propagation.In order to establish a simple,fast,efficient,efficient and effective system of rapid propagation and rapid propagation,the aim o f this system is to restore the 107 seeds of poplar in Europe and America,and has a positive guiding significance for the long-term promotion of young poplar.The main conclusions are as follows:1.The stem segments of two excellent single plants with top buds or axillary buds were used as explants.The stem segments with a long 1cm and each segment with a buds or axilla ry buds were harvested,and a large number of excellent single plant tissue culture seedlings were obtained at the end of the subculture medium.2.Orthogonal experiment was conducted to isolate and propagate the stem segments of good single bottle seedlings obtained by tissue culture.The results showed that the proliferati on and differentiation of 1/2MS+6-BA0.45mg L-1+NAA0.05mg L-1+GA1.0mg.L-11 medium was the best,with an average multiplication coefficient of 26.3.The shoot stem segment of a fine single plant seedling was transferred to the rooting medium to select the most suitable rooting formula.The experiment showed that the most id eal medium for rooting was 1/2MS+NAA0.02mg.L-1+IBA0.05mg.L-1,the rooting rate was100%,and the average root number was 5.9.4.In the process of detoxification of stem tip culture of poplar 107 in Europe and Ameri ca,the growth of stem tip was longer and browning was serious,so only 4 detoxified strains were successfully obtained,so the preservation rate of stem tip culture was low.By using?E LISA?detection technology to detect 4 strains,the results showed that the 4 strains of poplar tobacco embrittlement virus?TRV?were completely removed and the detoxification rate was100%,but only 3 of the 4 strains of poplar mosaic virus?PV-1?were removed and the detox ification rate was only 75%.5.In European and American poplar 107 with thermothermic treatment combined with stem tip detoxification,the growth of bottle seedlings was rapid during heat treatment,often accompanied by yellowing,shrinkage and landing.The longest tolerance time of the bottle w as 24D,the tip of the stem grew fast after heat treatment,and the browning phenomenon dec reased even no,and the preservation rate was higher.The results showed that the two viruses of TRV and PV-1 did not exist in the tested plants.The combination of heat treatment and st em apex could effectively remove all the two main viruses of TRV and PV-1,and the remov al rate was 100%.6.Western poplar 107 chemical combined with stem tip treatment detoxification,the re sults showed that when the addition of ribavirin 15mg.L-1,20mg.L-1,25mg.L-11 can also re move two viruses of TRV and PV-1,and two kinds of viruses can not be removed from 15m g.L-1.But during the experiment,the treatment of 15mg.L-1,20mg.L-11 and 25mg.L-11 ribavir in could inhibit the growth of the tube seedlings.The higher the concentration,the stronger t he inhibition effect.The inhibition effect was reduced or reduced to zero after the 15mg.L-1succeeding generations.And 20mg.L-11 and 25mg.L-11 ribavolazole sometimes severely inhib ited the growth of the bottle,so the preservation rate was very low15mg.L-11 is the optimum concentration for chemical treatment of poplar 107 in Europe and America.
