Priliminary Study On Intergeneric And Interspecific Hybridization Within Chrysanthemum In Broad Sense (â…¥)

The intergeneric and interspecific hybridization within chrysanthemum and its related genera were implemented in order to improve the germplasm resources of Chrysanthemum in broad sense, which also supply experimental evidence to identify the relationship among these genera.The tissue culture technology of Ajania purpurea was studied in this paper. Stem-segments of Ajania purpurea were used as explants and suitable media were selected respectively, the initial culture medium is MS+6-BA0.5mg-L-1+NAA0.01mg-L-1.; the propagation medium is MS+6-BA0.3mg-L-1+NAA0.05mg-L-1; the adventitious rooting medium prescription is1/2MS+IBA0.15mg·L-1.Based on the current germplasm resources, the intergeneric and interspecific crossings between Chrysanthemum and its relative genera were conducted, involving7genera (Chrysanthemum、 Elachanthemum、Hippolytia、Cancrinia、Opisthopappus、Argyranthemum、Nipponanthemum)17species, composing37combinations. After identified by morphological characters, chromosome ploidy observation by Flow Cytometry technology and nuclear ribosomal internal transcribed spacer (nrITS) gene sequence analysis, a kind of interspecific hybrid,a kind of intergeneric hybrid and a kind of trigeneric hybridization were confirmed to be real hybrids, and they are Chrysanthemum naktongense×C. argyrophyllum; Elachanthemum intricatum×Opisthopappus taihangensis; C. lavandulifolium×(Ajania achilloides×Opisthopappus taihangensis);also,the deduced hybrid Cancrinia maximowiczii×(Chrysanthemum naktongense×C. grandiflorum’Aifen’) that obtained by cooperative unit was identified to be true by Flow Cytometry technology.Intergeneric hybrid between Elachanthemum and Opisthopappus was obtained for the first time. The study provided direct evidence in researching the relationships among Chrysanthemum in broad sense.We got the trigeneric hybridization among Chrysanthemum,Ajania and Opisthopappus for the second time,which indicated the close relationship among the three genera.
