Seabuckthorn,also named Hippophae hamnoides.It is widely distributed in the temperate regions of Europe and Asia.Seen from the comprehensive analysis of the documents and materials reports,except the fruits of seabuckt-horn, the research of other parts of it just begins.In competitive sport,elimina-ting exhaustion and recovering strength are as important as sports training. How to put off the appearance of fatigue and advance the process of fatigue restoration before exercise is one of the hot issues of exercise science at pres-ent.This project applies the extract of the seabuckthorn(Hippophae rhamnoi-des ssp.sinensis) leaf to exhaustive swimming rat to research the regulating action and mechanism of the liver of the rat. It is expected this research can provide new ideas for eliminating exhaustion by theoretical basis for the exp-loitation of hippophae rhamnoides1. resource in Shanxi province.The biochemical parameters and cellular level,this paper builds a model about exhaustive swimming rat,by which the change of antioxidant enzymes of the liver of the exercise-induced fatigue rat is researched.Through this research,the effect and the regulating action that the seabuckthorn leaf extract caused to the free radical in the condition of exercise-induced fatigue is discussed.The effect and the regulating action that the seabuckthorn leaf extract for the liver cells is discussed by the research of the microstructure change of rat's liver cell level in sports fatigue.The preparation of the seabuckthorn leaf extract;Configuration seabuckt-horn leaf crude extracting samples;Identifying the bioactive compounds rut-in content by the high performance liquid chromatographic;Identifying the total phenolic content by Folin-Ciocalteu.Making a model of exhaustive sw-imming rats;Determining the most optimal does of seabuckthorn leaf extr-act.The testing of free radical level and other biochemical parameters.The pa-raffin sections of liver.Microscopical observation of liver cell structure.Research Result:Dissolving the seabuckthorn leaf extract in water and t-he extraction rate is:0.17g/1g dried Leaves.The result of rutin quantitative determination by HPLC in SBT leaf aqueous extracts is1.78μg mg-1.The evaluation of total phenolic content revealed that the extracts is rich in pheno-ls and contained75±5.3mg/g total phenols.Determine the most optimal does of seabuckthorn leaf extract by testing the swimming time of the rat.We make sure that the200mg/kg·BW is the best fittest does for the exhausted rat.The experiment shows that exhaustion sports have significantly increases in the activity of GSH-Px (P<0.05).Compared with the control group,the increase percentage is15.29%.But concentration of the MDA,the activity of SOD and the CAT have no significant influence.Taking seabuckthorn leaf extract has not important influence for the activity of MDA,SOD and the concentration of CAT(P>0.05).The overtraining movement has no significant improves in the activity of LDH (P>0.05).The seabuckthorn leaf extracts can reduce the activity of LDH (P<0.05).The content of hepatic glycogen is reduced signifi-cantly,Drinking seabuckthorn leaf extract can increase the content of Glc.The content of hepatic glycogen has significant difference in the groups of the control,but it has no significant influence in the activity of ALT and AST.In the research of the anatomic and histological structure of rats'liver,the norm-al group liver cells are perfect and complete.In exhausting exercise's group, the cells are incomplete.It has the size range of lipid droplets in bone cells;li-ver cells have varying degrees of necrosis.Here are seen the lipid vacuoles within hepatocytes.Lipid metabolism becomes dysfunctional.In seabuckthorn leaf extract supplement's group,the cells are incomplete and the lipid droplets disappear.There is no difference between the normal group and seabuckthorn leaf extract supplement group.As a important metabolite,it's a good antioxidant.It can eliminate the fr-ee oxygen radicals and block the degeneration of cells and cancers are preve-nted. Exhaustion swimming has played a very important role in content of glycogen and level of LDH in liver. The free radicals of rats become adaptab-le.The seabuckthorn leaf extract can protect the injury from the exhaustive swimming.It is required for normal metabolism in cells.Section observation provides the support from the cytology for the effect of seabuckthorn leaf extract has on the restoration of the exercise-induced exhaustion.
