Louhanguo is a species of siraitia in cucurbitaceae ,and it is a unique economic plant in Guangxi province.Widely used in Chinese medicine,drinks and condiments.The past is to use its fruit and leaves are usually discarded.In recent years it was to study the chemical composition of its leaves and found that it is rich in flavonoids.The main ingredient is quercitin and kaempferol.And found through in vitro that the flavons in siraitia grosvenorii leaf(FSGL) has a strong oxidation resistance.It is superior to synthetic antioxidant BHT.It is a good natural antioxidants. In this study, from the abandoned Luohanguo leaves extracted flavonoids, through high-intensity endurance training and made an exhaustive swimming sports injury model to study its anti-fatigue and stress on the movement of oxygen injury. The purpose of the development and utilization of the resources as well as its active role in flavonoid extract of the study provide the scientific basis.This study consists of two parts as follows:PartⅠ:Determine the dose of study. This study used a healthy 8-week-old male SD rats 48 and weighing 140-180 g. According to their weight,they were randomly divided into six groups, each with eight, namely: the control group , 100 mg group,200mg group,400mg group,800mg group,1600mg group. Incremental swimming training in two weeks, accurate records of the time immediately after exhaustive swimming,measure the plasma SOD activity and the content of MDA.The results show that, added a certain amount of FSGL could significantly prolong the time to swim to exhaustion, 200mg group to the most significant. 200mg group and in improving the plasma SOD activity and inhibition of MDA formation compared with the control group were significant differences (P﹤0.05) .And other effective dose group added there was no significant difference (P> 0.05). From the most effective point of view, will be 200 mg / kg weight of this dose as a formal experiment with the gavage doses. Part II:the flavons in siraitia grosvenorii leaf(FSGL) antioxidant protective effect on injury. This study used a healthy 2-3 month-old male SD rats 40 and weighing 180-220 g. According to their weight ,they were randomly divided into five groups, each with eight, namely: the quiet group, quiet Exhaustive group, quiet drug-taken group, exhaustive training group and exhaustive training + drug-taken group. Incremental swimming training four weeks after the quiet group were killed quiet and the rest of the group held its own weight 3 percent to exhaustive swimming,I accurate records of the time immediately after exhaustive broken blood, and from rat heart, liver and quadriceps were carried out biochemical tests and indicators of the morphology of the light microscope observation.The experimental results show that:(1)Use of the FSGL can significantly prolonged endurance training rats to swim to the duration of exhaustive, but there is no dose-dependent .This study showed that 200 mg / kg weight of the supplementary dose is most appropriate description of FSGL has strong anti-fatigue effect.(2) Use of the FSGL can improve after exhaustive swimming to rat plasma SOD, CAT, GSH-PX activity, decreased plasma MDA, BUN levels.Show that it can improve the body's antioxidant levels, reducing the consumption of protein during exercise, the increase in exercise capacity.(3) Use of the FSGL can improve after exhaustive swimming to rat heart, liver and quadriceps cells SOD, CAT, GSH-PX activity, reducing the content of MDA.Tip of the FSGL has strong anti-free radical antioxidant role.0(4) Use of the FSGL can be increased after exhaustive swimming to rat heart, liver and quadriceps-specific enzyme GOT, GPT, CK activity, decreased the plasma in the corresponding enzyme activity. Note the FSGL on the heart, liver and muscle cells are better protected, to a certain extent, guarantee the integrity of the cell membrane and mobility. (5) FSGL on different tissues in the same state of the antioxidant protective effect is difference. The experimental results showed that the liver and quadriceps stronger role in the protection of the heart tissue of the protective effect of the relatively weak.(6) Use of the FSGLcan increase swimming rats after exhaustive quadriceps Na+, K+-ATPase, Ca2+, Mg2+-ATPase and the ATPase activity. Tip of the FSGL to the maintenance of osmotic pressure inside and outside the cell membrane potential and transmembrane balance is of great significance, both inside and outside cells while maintaining Mg2+ Ca2+ homeostasis, and maintain higher energy supply, increase antioxidant capacity, delay fatigue campaign happen.(7) By optical microscopy observation of the tissues, found that use of FSGL the rat heart, liver and muscle cells more complete than the control group, greatly reduced the proportion of deformation. Tip of the FSGL on rat heart, liver and muscle cell membrane structure have some protective effect.(8) This study found that simply added FSGL or endurance training in the anti-fatigue in the body of the ability to protect against free radical damage to the body and the role of the regulation of energy metabolism Although also showed a certain role, but the two After the combined effect is that it can be markedly improved. Tip in the process of proper training FSGL may be added to improve the training of athletes with potential effect of the role.
