Object:To investigate the expression of SMAR1and CyclinDl in human stomach normal mucosa, malignant tumor by immunohistochemistry method, furthermore to study the relationship and clinic significance among the expression of SMAR1and CyclinDl in human stomach carcinoma, and analyzed the relationship between these protein expressions and the clinic pathological parameters, and the dependability among them.Materials and Methods:Collected surgical resection January1996and January2011the second affiliated hospital of kunming medical college and confirmed by pathology of gastric adenocarcinoma clinical material. randomly selected from73cases of gastric adenocarcinoma pathological specimens as the research object, its are primary tumor, and selected the46cases of cancer normal stomach tissue (from stomach cancer primary focal>5cm) as a control.These46patients with normal stomach were confirmed the infiltration of non-cancer cells or precancerous by histopa-theological study lesions.Expression of SMAR1and CyclinDl were ex-amined by S-P immunohistochemistry assays.The relationship between expression of SMAR1and CyclinDl in human stomach carcinoma were analyzed by SPSS17.0statistics software.Result:1. The expression of SMAR1were obviously lower in the carcinoma group than in the normol group. In46patients with normal gastric mucosal tissues in the positive expression rate was87.0%(40/46), and73cases of gastric cancer organizations in positive expression rate was45.0%(33/73). Gastric cancer and normol group SMAR1expression positive rate of significant differences(P<0.05),(respectively P<0.05). 2. The expression of CyclinDl were obviously higher in the carcinoma group than in the normol group. In46patients with normal gastric mucosal tissues in the positive expression rate was24.0%(11/46), and73cases of gastric cancer organizations in positive expression rate was61.6%(45/73).Gastric cancer and normol group CyclinD1expression positive rate of significant differences (P<0.0001)(respectively P<0.0001)3. The single factor analysis found that in the expression of gastric cancer organizations SMAR1with patients gender, age, tumor location, tumor size, histology, distant metastasis has nothing to with it. But it relevanted with tumor differentiation degree, infiltration depth, local lymph node metastasis, TNM staging (P<0.05). With the reduction of differentiation degree, the expression of the SMAR1declined. The high differentiation, differentiation, low differentiation in groups in turn positive expression rate is71.4%(5/7),42.9%(12/21),22.2%(10/45), the difference was statistically significant (X2=7.977, P<0.05). Along with the increase of the depth of the stomach wall infiltration, SMAR1positive expression rate declined. The T1, T2and T3, T4each group is in the positive expression rate83.3%(5/6),36.4%(4/11),28.0%(14/50),16.7%(1/6), the difference was statistically significant (X2=8.236, P <0.05). In the surrounding lymph node metastasis, NO, N1, and N2, N3positive expression rate was in turn51.2%(21/41),10%(1/10), and11.1(1/9),7.7%(1/13), the difference was statistically significant (X2=14.293, P<0.05).Visiblely surrounding lymph node metastasis group (NO) positive expression rate was significantly higher than the other group (N1, N2, N3). Along with the increase of TNM staging, the expression of the SMAR1declined. The Ⅰ, Ⅱ,Ⅲ, IV of the positive rate of expression was64.3%(9/14),40.6%(13/32),5.6%(1/18),11.1%(1/9), and the difference was statistically significant (X2=15.150, P<0.05).4. The single factor analysis found that in the expression of gastric cancer organizations CyclinDl with patients gender, age, tumor location, tumor size, histology, tumor differentiation degree has nothing to with it. But it relevanted with infiltrating depth, local lymph node metastasis, distant metastasis, TNM staging (P<0.05). Along with the increase of the depth of the stomach wall infiltration, CyclinDl expression is rising.The T1, T2and T3, T4each group of the positive expression rate was0%(0/6),27.2%(3/11),64%(32/50),83.3%(5/6). The difference was statistically significant (X2=14.302, P<0.05). In the surrounding lymph node metastasis, NO, N1, and N2, N3positive expression rate was39.0%(16/41),60.0%(6/10),88.9%(8/9),76.9%(10/13), and the difference was statistically significant (X2=11.019, P<0.05). Visiblely around lymph node metastasis (N3) group positive expression rate was significantly lower than the other group (NO, N1, and N2). In addition, the distant metastasis (M1) CyclinD1positive expression rate was100%(9/9), and no higher than the distant metastasis (MO)94%(31/64), and the difference was statistically significant (X2=8.469, P<0.05). Along with the increase of TNM staging, CyclinD1of expression was rising gradually. The I, II, III, IV of the positive expression rate was7.1%(1/14),50.0%(16/32),77.8%(14/18),100%(9/9), and the difference was statistically significant (X2=24.394, P<0.05).5. Spearman rank correlation analysis with SMAR1and CyclinDl the correlation between the two indexes. The results showed that the expression of SMAR1and CyclinD1have negative relationship with each other in the cancer of the stomach (respectively P<0.01).Conclusion:1. The expression of SMAR1was obviously lower in the carcinoma group than in the normol group. The expression of SMAR1have relationship with the degree of differentiation degree, infiltration depth, local lymph node metastasis, TNM staging of stomach malignant tumor.2. The expression of CyclinD1was obviously higher in the carcinoma group than in the normol group. The expression of it have relationship with it infiltration depth, local lymph node metastasis, distant lymph node metastasis, TNM stagingof stomach malignant tumor.3. There was negative correlation between the expression of SMAR1and CyclinDl in the gastric carcinoma tissues.4. The cancer of the stomach SMAR1protein and organization CyclinDl protein expression differences may and stomach cancer occurrence, development process closely related, both the abnormal expression in the evolution of the gastric cancer may play a different role; In the stomach cancer SMAR1expression in the fall and CyclinDl express, the rise of the gastric cancer patients may help screen, still can be in in the future for clinical diagnosis, prognosis of gastric cancer early and targeted therapy provides important theoretical basis.
