OBJECTIVE:To study the relationship between leptin receptor (Lepr) gene polymorphism and hypertension and obesity in han population.METHORDS:Select han hypertension283cases hospitalized patients,choose153cases during the same period as normal control group. According to body mass index (BMI)≥28kg/m2, hypertension group is classified as patients of hypertension with obesity group162cases and121of patients of hypertension without obesity group. Using polymerase even response--restriction fragment length polymorphism (PCR-RELP) method for determining Gln223Arg(rs1137100) and Lys109Arg(rs1137101) Lepr gene polymorphism, ELISA determination leptin levels in blood. Carotid intima-media thickness (CIMT) were measured by ultrasonography (American PHILIP-IE33, L11-3,11MHz) in all subjects.RESULTS:The EH group had a higher BMI, blood pressure, CIMT and leptin level (P<0.05) compared with contorl group..The genotype frequency of Gln223Arg variant in leptin receptor gene in the case group and the control group had a significant difference (P<0.05).The genotype frequency of Lys109Arg variant in leptin receptor gene in the case group and the control group had no significant difference (P>0.05). Gln223Arg alleles Lepr gene frequency of A showed significant difference between the patients of hypertension with obesity and the patients of hypertension without obesity(P=0.000). and patients of hypertension with obesity group has A higher BMI and CIMT.Lepr gene Lys109Arg frequency alle A and alle G showed no significant difference in the patients of hypertension with obesity (P>0.05)CONCLUSION:Lepr gene polymorphism Gln223Arg was possibly associated with hypertension and obesity in Han population. And A allele is the risk factor to atherosclerosis in the patients of hypertension with obesity group. Lys109Arg Lepr gene polymorphism has no obvious related to hypertension and lipid metabolism disorders.
