| ObjectiveIdiopathic pulmonary fibrosis (IPF) is a kind of disease characterized by diffuse alveolitis and alveolar structure destruction which finally leading to fibrosis, its lung function is characterized with limitory ventilation interference, and diffusion restruction. According to the survey of epidemiology, the annual percentage rate of this disease is about 0.7-1%, which people commonly aged between 50-70 years old, whose livability are less than 3 years. And it is also indicated that with the age developing, the incidence and mortality of this disease is increasing. Because of no specific drug and dissatisfactory effects, IPF is difficult to treat. At present, the urgent question affronting the medical science field is to study the pathogenetic mechanisms and seek effctive drug for IPF.The research show that cucumin possess widespread pharmacological action in recently years, for example anti-inflammatory, anti-oxidation, anti-ischemic, anticancer, inhibiting hepatic fibrosis. To discuss the preventing and treating effect of cucumin on idiopathic pulmonary fibrosis, rat model induced by bleomycin (BLM) was adopted. This thesis describes the effect and mechanisms of cucumin on IPF from aspects of integral and molecular level, which can provides experimental foundation for more exact and scientific therapy.Methods&Results1 Experimental study on the effect of cucumin on IPF.The effect of cucumin on extracellular matrix (ECM) and lung function144 male Sprague-Dawley rats were randomly divided into 6 groups (24 rats in each group). Rats in the model control group, positive medicine group, and high, moderate and low cucumin groups were injected with a single dose of bleomycin by trachea, and rats in sham-model control group with same volume normal saline. 1 day after the injection, oral administration of drug daily until they were killed 1 day before. Cucumin solution of different dosages (200mg,100mg,50mg·kg-1·d-1) was respectively given to rats in the high, moderate and low cucumin group by gavage, while equal volume of normal saline was given to those in the sham-model control group and model control group, and an equal volume of prednisone(0.56mg·kg-1·d-1) was saline was given to those in positive medicine control group.On the 7, 14, 28 day, the lung function were test in fibrotic rats, the concentration of hydroxyproline in lunghomogenates and the levels ofⅢ-collagen,Ⅳ-collagen, laminin and hyaluronicacid in serum were determined.
