Obsject Observing the effect of common turmeric n-butanol extraction on the pancreas fibrosis in chronic pancreatitis rat induced by Diethyldithiocarbamic acid sodium salt(DDC),we explore the mechanism of that.Methods 66 male, health and clean Sprague-Dawley rats, divided randomly into the group of blank control (group A,n=12), the group of model(group B,n=18), the group of high dose common turmeric extraction by n-butanol (group C,n=18), the group of low dose common turmeric extraction by n-butanol (group D,n=18).The rats model of pancreas fibrosis were made with intraperitoneal injection DDC:The first week to the fourth week,750mg/kg DDC were injected into the abdominal cavity twice a week, the fifth week to the ninth week, 750mg/kg DDC were injected into the abdominal cavity once a week, at the tenth week, we can get pancreatic fibrosis model rat. After mading model one week, the rats of group A were given normal saline(1 ml/kg) by gavage after one week normally fed;the rats of group B given normal saline(1 ml/kg) by gavage every day. the rats of group C given high dose common turmeric n-butanol-extraction(equivalent to the crude herb 5g/kg) by gavage every day, the rats of group D given low dose common turmeric n-butanol-extraction(equivalent to the crude herb 1.25g/kg) by gavage every day, at the same time,the rats were mading model. After ten weeks, anaesthetized the rats,drew blood from abdominal aorta,got pancreas tissues.observed the gross morphology of pancreas, calculated the score of injury in HE staining, measured the aera of collagen fiber in MASSON staining, observed the a-SMA expression by immunohistochemistry, measured the level of Hyp in serum by ELISA. At last analysis the correlation.Results①General situation and pancreas gross morphology:In control group, the general situation and pancreas gross morphology were well; In model group, the general situation were worse, the injury of pancreas were severe.Compared with the model group, the general situation and pancreas gross morphology of high dose common turmeric n-butanol-extraction group and low dose common turmeric n-butanol-extraction group were improved obviously.②HE staining:In control group, the organization structure of pancreas were normal, the score of injury in control group was(0.08±0.29); In model group, the injury of organization structure in pancreas were obvious, the score of injury in model group was(4.56±1.13). the injury in intervention group were milder than model for all (P<0.05 for all), the score of injury in high dose common turmeric n-butanol-extraction group was (0.81±0.83),the score of injury in low dose common turmeric n-butanol-extraction group was (0.93±0.96).Compared with high dose common turmeric n-butanol-extraction group,the injury of pancreas in low dose common turmeric n-butanol-extraction group was similar,and the difference was not significant(P>0.05).③MASSON collagen fiber staining:In control group, the distribution of collagen fiber were aroud the vascular, and the percentage of collagen fiber in control group was (1.83±1.02)%.In model group,the collagen fiber was widely and densely distributed between the pancreatic lobules, and the percentage of collagen fiber in model group was(13.46±5.58)%, the percentage of collagen fiber in high dose common turmeric n-butanol-extraction group was (3.34±1.48)%. The percentage of collagen fiber in low dose common turmeric n-butanol-extraction group was(3.51±2.19)%,the collagen fiber in intervention groupgroup were much less than model group for all (P<0.05 for all).Compared with high dose common turmeric n-butanol-extraction group,the collagen fiber area of pancreas in low dose common turmeric n-butanol-extraction group was similar,and the difference was not significant(P>0.05).④The expression of a-SMA by immunohistochemistry:In control group,the expression of a-SMA were only aroud the vascular; In model group,the expression of a-SMA was more intensive,the distribution of brown granules was wide;The expression of a-SMA of pancreas in intervention groups (group C or D) were much less than the model group for all(P<0.05 for all).Compared with high dose common turmeric n-butanol-extraction group, the expression of a-SMA of pancreas in low dose common turmeric n-butanol-extraction group was similar, and the difference was not significant (P>0.05)。⑤The serum Hyp level by ELISA:The serum Hyp level of rat in control group was lower;The serum Hyp level in model group was higher.(34.03±15.87)μg/L; The serum Hyp level of rat in high dose common turmeric n-butanol-extraction group was(23.50±12.67) P g/L,the serum Hyp level of rat in low dose common turmeric n-butanol-extraction group was (22.66±10.65)μg/L,the serum Hyp level in intervention group were much less than model group for all (P<0.05 for all).Compared with high dose common turmeric n-butanol-extraction group,the serum Hyp level of rat in low dose common turmeric n-butanol-extraction group was similar, and the difference was not significant (P>0.05)。Correlation analysis results:the aera of collagen fiber,the a-SMA expression and the level of Hyp in serum had a positive correlation with the score of injury in HE staining,(r were respectively 0.819,0.910,0.468, P all<0.01); the a-SMA expression and the level of Hyp in serum had a positive correlation with the aera of collagen fiber, (r were respectively 0.736 (P<0.01),0.340 (P<0.05));the relationship between the a-SMA expression and the level of Hyp in serum was positively correlated (r was 0.354, P<0.05).Conclusion The extraction by n-butyl alcohol in common turmeric can effectively relieve the fibrosis of pancreas in chronic pancreatitis rat induced by DDC. The mechanism of that may have a correlation with the inhibiting the expression of a-SMA and reducing the aera of collagen fiber in pancreas...
