Objective:To observed the expression of aquaporin 2(AQP2) and aquaporin 3(AQP3) in nephritic syndrome (NS) rats induced by Aadriamycin and to explore the therapeutic mechanism of Liyu Chiciaodou Tang on NS.Methods:Total of 50 health adult Wistar rats were randomly divided into five groups consisting of 10 rats respectively: normal group (group N), NS group (group M), The rats of group NS were treated with Fosinopril (group F), group NS with lower amounts Liyu Chixiaodou Tang (group D), group NS with higher amounts Liyu Chixiaodou Tang (group G), received 9ml/kg supplementation, total intervention 3 weeks. The urinary protein excretion of rats was determined once before and every three days after injection adriamycin (ADR). The levels of serum total protein (TP), plasma albumin (ALB), serum creatinine (Scr), blood urea nitrogen (BUN), sodium, postassium and chlorine were measured after 3 week of treatment. The pathological changes were observed by light microscope, and the expression of AQP2 and AQP3 were measured by immunohistochemistry assay.Results:(1) 12h urinary albumin excretion:there was no significant difference before and after established nephropathy models among these groups (before:F=0.073, P=0.79; after:F=0.387, P=0.76), but there were statistically difference compared with group N after established nephropathy models(P<0.01), which suggested the NS models established successful. The urinary protein excretion in group F,G,D decresed at the first 9 days, (P<0.01, compared with group M). (2) The TP and ALB of group G,D,F were increased(P<0.05,compared with group M). group G was better than group F(P<0.05). (3) The serum sodium, chloride of group G,F were decreased(P<0.05,compared with group M). (4) electron microscope: There was no change in glomerular's appearance and stryctures of group N. Group M's hyperplasia of glomerulus,mesangial cell and basement membrance, with the infiltration of inflammatory. In group G,D,F, the pathological changes was lessed than group M. (5) Immunohistochemistry assay: In group M, protein expression of AQP2 and AQP3 had decresed in renal tubules, and significantly higher than group N(P<0.05). Compared with group M, protein expression of AQP2 and AQP3 had been remarkably decreased in group G,D,F (P<0.05).Conclusion:In adriamycin nephropathy rats, Liyu Chixiaodou Tang could decrease urinary albumin excretion, increase TP and ALB, decrease soddium and chloride, and relieve the pathological changes, decrease the expression of AQP2 and AQP3 in renal tubules.
