Objective:To observe the expression of podocyte’s Nephrin and CD2AP in adriamycin nephritic rats, and to discuss the effects of Liyu Chixiaodou Tang on them.Methods:50male Wistar rats in SPF grade were randomly divided into two parts,10for normal group (N group) and40for pre-model group. The rats of pre-model group were injected with adriamycin(6.5mg/kg)via tail vein for one time to copy the kidney disease model, and then they were randomly divided into model group (M group), fosinopril group (F group),Liyu Chixiaodou Tang low-dose group(DD group), Liyu Chixiaodou Tang high-dosage groups (DG group),each group got10. F group, DD group, DG group were treated with appropriate medicine or soup once a day for7weeks. Then the specimens were collected. The rats’12h urinary protein, albumin, total protein were detected, the renal’s pathological changes were detected by optical microscope, the ultrastructural lesions were examined by electron microscopy. The expression levels of Nephrin and CD2AP were examined by immunohistochemistry.Result:①Compared with the N group’s, the other groups’12h urinary protein were significantly increased(p<0.05), especially for the M group’s; Compared with the M group’s, the treated groups’12h urinary protein were all reduced in varying degrees, but the differences were not statistically significant.②Compared with the N group’s, the M group’s expression of albumin and total protein were decreased(p<0.05), while the treated groups’only got the albumin decreased significantly(p<0.05), their total protein also decreased, but with no statistically significant; Compared with the M group’s, the treated groups’expression of albumin and total protein were all increased, but only the DG group’s were statistically significant(P<0.05).③Optical microscope:There was no abnormal changes in the N group’s kidney tissue; The M group’s glomerular capillaries and glomerular capsule were stuck together, the thbular lumen was filled with large numbers of protein casts, the renal interstitial was significantly infiltrated by lymphocyte, some of them also became fibrosis; The treated groups’pathological changes were all reduced than the M group’s. Electron microscopy:Glomerular podocytes, basement membrane and endothelial cells were normal in the N group; The M group’s podocyte foot processes were fused extensively, some of them even disappeared, the basement membrane’s boundaries were blurred, the fenestia of the endothelial cell was disappeared; The treated groups’ultrastructural pathological changes were all reduced than the M group’s.④Compared with the N group’s, the other groups’expression of Nephrin and CD2AP in glomerular were decreased(P<0.05), especially for the M group’s; compared with the M group’s, the treated groups’expression of Nephrin and CD2AP in glomerular were increased(P<0.05), however the differences between the treated groups’expression were not statistically significant.Conclusion:The abnormal expression of Nephrin and CD2AP is an important molecular mechanism during the disease progression. Liyu Chixiaodou Tang can improve their albumin and total protein, and can protect the glomerular filtration barrier though improving the expression of the Nephrin.and CD2AP. |
