| Objective To investigate the association between single nucleotide polymorphism G/A in the promoter of leptin gene and abdominal obesity cerebral infarction(CI) in Chinese Han people of Qingdao city.Methods The genotypes of leptin were detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) methods in 216 patients with cerebral infarction and 182 cases of unrelated healthy people. The two groups were respectively divided into with and without abdominal obesity groups, which were divided into 4 subgroups and were carried out polymorphism analysis.Result (1)The distribution genotype was in Hardy-Weinberg. (2)The genotype frequencies of GG,GA and AA were 0.1991,0.5417 and 0.2593, the distribution frequencies were 0.0714,0.4231 and 0.5055 in the control group. The difference was statistically significant(X2= 13.311, P<0.001). The relative risk suffering from CI with GG was 3.231 times of those with genotype GA+AA[OR(GG/GA+AA)=3.231,95%CI:1.677-6.224]. The allele frequencies of G and A were 0.4699 and 0.5301 in CI group and they were 0.283 and 0.717 in control group. The difference was statistically significant(X2=16.429, P<0.001). The CI morbidity of G allele carriers is 1.859 times of A allele carriers[OR(G/ A)= 1.859,95% CI:1.375~2.513]. There is no obvious difference of genotype frequency between abdominal obesity group and non-abdominal obesity group (P>0.05).Conclusion Leptin gene polymorphism G-2548A is significantly correlation with the CI and it is not associated with abdominal obesity. G allele of leptin may be a genetic factor that may contribute to individual susceptibility for CI. |
