Objective To observe the effects of Cyprinus carpio decoction on the expression of aquaporin-1, aquaporin-2 with adriamycin-induced nephropathy rats in order to provide experimental basis for the treatment of nephrotic edema.Methods Total of 50 Wistar rats were randomly divided into normal group (group N), model group (group M), fosinopril group (group F), Cyprinus carpio decoction with high dose group (group YG), and Cyprinus carpio decoction with low dose group (group YD). Nephropathy models were established by injecting adriamycin through tail vein. Urinary protein excretions in 12h, serum albumin, total serum protein, triglyceride, total cholesterol, triglyceride, total cholesterol, blood urea nitrogen, creatinine, serum sodium and potassium were measured. The pathological changes and the expression of AQP1, AQP2 in rat kidneys were respectively detected by HE stain and immunohistochemiscal assay.Result①The urinary protein excretion in 12h had no significant differences among the groups before modeling (P>0.05). All model rats emerged proteinuria after modeling, there was significant difference compared with group N (P<0.05). Urinary protein excretion of group M increased significantly with time. Urinary protein excretion of group YG, group YD and group F increased slowly and improved with different degrees.②Compared with group N, serum albumin and total protein of each group were significantly lower (P<0.01). Serum albumin and total protein of group F, group YD and group YG increased (P<0.05) compare with the group M, but only group YG had significant difference (P<0.01). After the model, the serum total cholesterol and triglyceride levels increased in varying degrees (P<0.01), but the group M and drug intervention groups were not statistically significant (P>0.05). Urea nitrogen and creatinine showed no differences among groups (P>0.05).③Renal glomerulus, renal tubular interstitial ormal and small blood vessels were normal in group N. Renal glomerular capillaries congestive of group M rats were expansion. Glomerular mesangial cells and the basement membrane were diffuse hyperplasia, inflammatory cell infiltration. Glomerular mesangial cells and the basement membrane of group F, group YG, group YD reduced proliferation, less inflammatory cell infiltration.④The expression of AQP1 in each group within the proximal tubule and the cell membrane in thin section of medullary loop was significantly increased compared with group N (P<0.01) after modeling. The expression of group YG, group YD and group F was significantly higher than group M (P<0.01). AQP2 was expressed in the renal distal tubules and luminal side of collecting duct principal cells. Compared with Group N, the expression of AQP2 in the other groups increased, the difference was significant (P<0.01). The expression of AQP2 in group YG, group YD and group F decreased compared with group M (P<0.05). The expression of group YG were significantly lower (P<0.01) than group M.Conclusion Cyprinus carpio decoction could decrease the expression of AQP1, AQP2 in rat kidneys, alleviate pathological lesion with adriamycin-induced nephropathy rats, and reduce urinary protein excretion of rats.
