Objective:To study the protective effect of grape procyanidins on alcoholic liver injury in mice.Method:50 mice were randomly divided into 5 groups:normal group of control, the group of liver injury induced by alcohol,the groups of procyanidins with low doses(50mg/kg), middle doses(100mg/kg) and high doses(200mg/kg). The mice were orally given the dose of 4g.kg bw-1 ethanol and different doses(50,100,200 mg/kg) of GPC except the normal control group with distilled water once a day. Observed mice growth and the mice were killed after 30 days. Then the liveness of alanine aminotransferase (ALT),asparate aminotransferase (AST),total protein, total cholesterol and triglycerides in serum were detected. The levels of PCNA and Bcl-2 protein expression in the cell were measured by immunohistochemistry method. The proliferation activity of the hepatic cells was determined by MTT assay.Results:1.Results of liver weight,body weight and liver index Compared with the normal control group, the liver weight and liver index in ethanol group were significantly increased (p<0.05). The liver weight and liver index of GPC mid and high dose group were significantly lower than that of the ethanol group(p<0.05). 2.Results of serum ALT, AST, and total protein, total cholesterol, triglyceride assay Compared with the normal control group, the liveness of serum ALT,AST and triglyceride in ethanol group were significantly increased (p<0.05). Compared with the ethanol control group, ALT, AST and triglyceride levels in GPC mid and high dose group were decreased significantly (p<0.05). There was no significant difference between normal control group and GPC high dose group p> 0.05). The serum total cholesterol, total protein level of the normal control group, GPC mid and high dose group were significantly higher than that of ethanol group (p<0.05).3. Results of PCNA protein expression in liver cells The proliferation index of PCNA protein of the high-dose GPC group was 39.26%, and that of the ethanol group was 9.5%.The difference between the two groups was significant(p<0.05). Compared with the GPC low dose group, PCNA proliferation index of GPC mid and high dose group were increased significantly (p<0.05).4. Results of Bcl-2 protein expression in liver cells The expression rates of Bcl-2 protein of the ethanol group was 9.22%, significantly lower than those of GPC mid and high dose group(p<0.05).And the differences between GPC low dose group and GPC high and mid dose group were significant (p<0.05).5. Results of liver cell proliferation The proliferation activity of GPC high-dose group was 0.423±0.125, significantly higher than those of the ethanol control group and GPC low-dose group (p<0.05),but compared with the normal control group there was no significant difference (p> 0.05).Conclusion:GPC had inhibitory effect on the decrease of proliferation activity and Bcl-2 protein expression.It had protective effect on liver injury induced by ethanol in mice.
