Objective Grape procyanidins (GPC) are natural polyphenols extracted fromgrapes. It was reported that GPC have a lot of bioactivities such asantioxidation,antiradical and anti-tumor effects. In this study,the effects of grapeprocyanidins on the proliferation of breast carcinoma cells and normal breast cellswere observed in vitro and vivo.Methods1. Vitro ExperimentThree types different human breast cells, that is, breast carcinoma cells, breastcarcinoma cells treated with cyclophosphamide and normal breast cells, were culturedwith different concentration of GPC. MTT assay was utilized to investigate theproliferation of different breast Cells.2. Vivo Experiment40 mice who were inoculated EMT-6 breast carcinoma cells were randomlydivided into 4 groups, carcinoma comparison group, low-dosage GPC group,middle-dosage GPC group, high-dosage GPC group. Different dose of GPC wasgiven by mouth to the mice in all GPC groups. The mice in low-dosage GPC group,middle-dosage GPC group and high-dosage GPC group were gavaged respectivelywith 10mg/kg,100 mg/kg and 200 mg/kg body weight GPC for 14 days, and onlywater was gavaged to the mice in Carcinoma comparison group. The proliferation ofbreast carcinoma cells was determined by the same assay,and the PCNA and p53expression of mice breast carcinoma cells was measured by immuchistochemistry.Results1. Vitro ExperimentWith the increasing dose of GPC,the proliferation of breast carcinoma cellsdecreased, but the inhibition ratio rised. For the normal breast cells, the proliferationrised with the increasing dose of GPC. The inhibition action of grape procyanidinson proliferation of breast carcinoma cell depended on concentration.Compared with the cyclophosphamide group and carcinoma comparison group,the proliferation of the cyclophosphamide plus middle-dosage GPC group or thecyclophosphamide plus high-dosage GPC group depressed obviously, thedifferences above were significant in statistics (p<0.05).2. Vivo ExperimentThe proliferation of carcinoma comparison group, middle-dosage GPC group, high-dosage GPC group was 1.098±0.167, 0.714±0.037 and 0.881±0.120respectively, the difference above were significant in statistics (p<0.05).The expression rates of PCNA and p53 of the carcinoma comparison group were 72.28%and 37.22% respectively, and those of the high dose GPC group were 28.32% and 7.46%.Thedifferences between the two groups were significant (p<0.01).Conclusion1. GPC can suppress the proliferation of the breast carcinoma cells.2. GPC can promote the proliferation of normal breast cells.3. GPC and cyclophosphamide present coordinate repression to the proliferation ofbreast carcinoma cells.4. This study will offer scientific theory to treat and prevent some breast carcinoma...
